Biochemische Untersuchung von Hyperforin-verwandten Prenyltransferasen, die in Saccharomyces cerevisiae experimiert wurden

Hypericum perforatum, known as St. John´s wort, is one of the most famous and best investigated medicinal plant. Hyperforin identified as the major constituent responsible for the main pharmaceutical effects (the treatment of mild to moderate depression) of the extract. It is light, oxygen, and temperature sensitive hydrophobic acylphloroglucinol derivative with several prenyl and one geranyl side chains. However, its biosynthetic pathway is not yet completely understood. As proposed, the biosynthesis of hyperforin starts with the formation of the core skeleton, phlorisobutyrophenone (PIBP), catalyzed by a type III polyketide synthase. The subsequent decoration of PIBP with prenyl and geranyl side chains is catalyzed by aromatic membrane-bound prenyltransferase (PT) enzymes. The aim of this project was the identification and characterization of enzymes involved in the hyperforin biosynthesis. In this study, four consecutive enzymes encoding an acyl-CoA ligase (HpIBCL), an isobutyrophenone synthase (HpIBS) and two consecutive PTs were functionally expressed in S. cerevisiae. The first step was the de novo biosynthesis of the hyperforin skeleton PIBP. Therefore, cDNAs encoding HpIBCL and HpIBS were cloned from H. perforatum. Feeding isobutyric-acid to S. cerevisiae harboring either episomal or genome-integrated HpIBCL and HpIBS resulted in the detection of intracellular and extracellular PIBP. Totally twelve PTs were individually expressed in S. cerevisiae, one PT (HpPT2) being identified to prefer the geranylation of acylphloroglucinol-derivatives PIBP/2-MBP and GPP as the major substrates. A second PT (HpPT6) catalyzed the addition of one prenyl group to the HpPT2 product forming the hyperforin precursor molecule 3-geranyl-5-prenyl-PIBP. The biochemical properties of both PT enzymes were further characterized. Both PT enzymes were found to show typical characteristics of plant derived PTs such as divalent cation dependency, broad substrate specificity, and alkaline pH optimum. KM values for 2-MBP and PIBP were 85 ± 15 µM and 124 ± 19 µM for HpPT2, respectively. KM values for the HpPT6 substrates 3-G-PIVP, 3-G-2-MBP and 3-G-PIBP were 78 ± 8.6, 73 ± 10, and 181 ± 218, respectively. These results might confirm the identification of the first PT enzyme (HpPT2) involved in the hyperforin biosynthesis. Despite its questionable biochemical properties of HpPT6, the in vitro ability to catalyze the biosynthesis of a hyperforin precursor is a promising approach.Hypericum perforatum (echtes Johanniskraut) ist eine der bekanntesten und am besten untersuchten Heilpflanze der Welt. Sie wird hauptsächlich zur Behandlung von leichten bis mittelschweren Depressionen eingesetzt, wobei Hyperforin als Hauptinhaltsstoff für die Wirkung verantwortlich gemacht wird. Es ist ein licht-, sauerstoff- und temperaturempfindliches hydrophobes Acylphloroglucinol-Derivat mit mehreren prenyl- und einer geranyl-Seitenkette. Die Biosynthese ist bisher noch unbekannt. Die vorgeschlagene Biosynthese beginnt mit der Bildung des Phlorisobutyrophenon (PIBP), gebildet durch eine Typ-III-Polyketidsynthase. Anschließend katalysieren aromatische Prenyltransferasen (PT) die Anknüpfung von isoprenoid-Seitenketten an das PIBP. Ziel dieses Projektes war die Identifizierung und Charakterisierung von Enzymen aus H. perforatum, welche an der Hyperforin-Biosynthese beteiligt sind. In dieser Studie konnten 4 konsekutive Enzyme, eine Acyl-CoA-Ligase (HpIBCL), eine isobutyrophenone-synthase (HpIBS) und zwei aufeinanderfolgende PTs in S. cerevisiae funktionell exprimiert werden. Der erste Schritt war die Biosynthese der Hyperforin Grundstruktur (PIBP). Hierfür wurden zwei cDNAs, jeweils eine für HpIBCL und HpIBS aus H. perforatum kloniert und entweder Plasmid basierend oder Genome integriert exprimiert. Anschließend konnte durch Fütterungsversuchen mit Isobuttersäure das PIBP Produkt intrazellulär und extrazellulär nachgewiesen werden. Insgesamt wurden zwölf PTs in S. cerevisiae exprimiert. Hierbei wurde eine PT (HpPT2) identifiziert, welche die Acylphloroglucinol-Derivate PIBP, 2-MBP sowie GPP bevorzugt. Eine zweite PT (HpPT6) konnte identifiziert werden, die eine weitere prenyl-Seitenkette an das HpPT2 Produkt anheftet und so das Hyperforin-Vorläufermolekül 3-Geranyl-5-prenyl-PIBP bildet. Zusätzlich wurden die biochemischen Eigenschaften beider PTs bestimmt. Beide wiesen typische Eigenschaften pflanzlicher PTs auf, wie z.B. Kationenabhängigkeit, breite Substratspezifität und alkalischer pH-Optimum. KM - Werte für 2-MBP (85 ± 15 µM) und PIBP (124 ± 19 µM) für HpPT2 konnten bestimmt werden sowie für die HpPT6-Substrate 3-G-PIVP (78 ± 8,6), 3-G-2-MBP (73 ± 10) und 3-G-PIBP (181 ± 218). Die erhaltenen Ergebnisse könnten die Identifizierung der ersten PT (HpPT2) bestätigen, welches an der Hyperforin-Biosynthese beteiligt ist. Die biochemischen Eigenschaften von HpPT6 sind noch fraglich, jedoch ist das gebildete Produkt ein vielversprechende Hyperforin-Vorstufe
