Studies of the structure and function of selected polyketide synthases

Typ-III-Polyketidsynthasen (PKS) katalysieren in Pflanzen die Bildung wichtiger Sekundärstoffe, die pharmakologisch interessante und chemisch kompliziert aufgebaute Wirkstoffe darstellen. Da die Komplexizität der Strukturen eine chemische Synthese häufig unmöglich macht, liegt in der Erforschung der Biosynthese dieser Stoffklasse und der daran beteiligten Enzyme ein großes Potential für neue Therapiemöglichkeiten. Die Biphenylsynthase (BIS1, Sorbus aucuparia, Rosaceae) katalysiert die Bildung von 3,5-Dihydroxybiphenyl, welches das Grundgerüst des Aucuparins darstellt. Die Kristallisierung dieses Enzyms gelang nach Aufreinigung über Ni-NTA-Agarose bei einem mit TEV-Protease geschnittenen Konstrukt in unterschiedlichen Kristallisationspuffern. Auf die gleiche Art konnte die Benzophenonsynthase (BPS, Hypericum androsaemum, Hypericaceae), welche die Bildung von 2,4,6-Trihydroxybenzophenon katalysiert, kristallisiert werden. 2,4,6-Trihydroxybenzophenon ist das Grundgerüst von Phytoalexinen des Johanniskrauts, dessen Extrakte wegen ihres Hyperforin-Gehalts weite Anwendung als Antidepressivum finden. Für eine Mutante dieses Enzyms gelang außerdem die Beweisführung der zuvor aufgestellten Inhibierungshypothese durch ein enzymatisch gebildetes Produkt. Mit Hilfe von Enzymtests wurde außerdem die Phenylphenalenonsynthese in Wachendorfia thyrsiflora (Haemodoraceae) untersucht. Für die beiden Hauptsubstrate Cinnamoyl-CoA und p-Cumaroyl-CoA folgt die enzymatische Reaktion einer sigmoiden Kinetik, wobei die Sättigungskonzentrationen bei 150 µM bzw. 700 µM lagen. In nicht-denaturierenden Polyacrylamidgelen konnte gezeigt werden, dass die zwei Polyketidsynthasen vom Typ III (WtPKS1 und WtPKS2), deren cDNAs aus Zellkulturen der Pflanze isoliert und exprimiert wurden, beim Aufbau des Bisdesmethoxycurcumins wahrscheinlich als Komplex zusammenarbeiten. Eine solche Komplexbildung wurde bisher für keine Typ III PKS beschrieben.Type III polyketide synthases (PKSs) catalyse the formation of important plant secondary metabolites, which constitute pharmacologically interesting and chemically challenging agents. The crystallization of this enzyme was successful in various crystallization buffers after purification via Ni-NTA-agarose using a construct cleaved by TEV-protease. Likewise, benzophenone synthase (BPS, Hypericum androsaemum, Hypericaceae) which catalyses the formation of 2,4,6-trihydroxybenzophenone was crystallized. 2,4,6-Trihydroxybenzophenone is the skeleton of phytoalexins produced by St. John’s wort (Hypericum perforatum), extracts of which are widely used as antidepressants due to their hyperforin content. Furthermore, the inhibition hypothesis postulated previously for a mutant of this enzyme was successfully proven via an enzymatically formed product. In addition, the phenylphenalenone synthesis in Wachendorfia thyrsiflora (Haemodoraceae) was examined using enzyme assays. For the two main substrates cinnamoyl-CoA and p-coumaroyl-CoA, the enzymatic reaction followed a sigmoid kinetic with the concentration of saturation being 150 µM and 700 µM, respectively. In non-denaturating polyacrylamide gels, the two type III polykedide synthases (WtPKS1 and WtPKS2), cDNAs of which had previously been isolated and expressed from cell cultures, were found to cooperate as a potential complex in the biosynthesis of bisdesmethoxycurcumin.
