Hyperforin-Biosynthese: Charakterisierung und Reinigung einer Prenyltransferase aus Hypericum calycinum-Zellkulturen

Hyperforin is a broad-band neurotransmitter reuptake inhibitor from Hypericum perforatum (St. John's wort). Extracts from this medicinal plant are widely used for their antidepressant activity. Hyperforin is a polyprenylated phloroglucinol derivative. The enzyme which catalyzes the first prenylation step in hyperforin biosynthesis was detected in cell cultures of H. calycinum grown in BDS medium in the dark. These cell cultures thus turned out to be a valuable in vitro system for studying hyperforin biosynthesis. The transferase is soluble and its activity dependent on a divalent cation, preferably ferrous (Km = 3.8 mM). The combination of these two properties is unusual for aromatic prenyltransferases. The preferred prenyl donor for the enzyme was DMAPP and the preferred prenyl acceptor phlorisobutyrophenone. The apparent Km values for dimethylallyl diphosphate and phlorisobutyrophenone were 0.46 and 0.52 mM, respectively. The enzyme exhibited a broad pH optimum between 6.5 and 9.3. The accumulation of hyperforins in H. calycinum cell cultures was preceded by an increase in dimethylallyltransferase activity, with the maximum specific activity being 3.6 μkat/kg protein. The enzyme was purified using DEAE and Mono-Q anion exchange chromatography, hydroxylapatite adsoption chromatography, hydrophobic interaction chromatography and gel filtration. On a calibrated gel filtration column, the relative molecular mass (Mr) of the dimethylallyltransferase was 63 000. In SDS-PAGE, the purified protein gave two closely spaced polypeptide bands with Mr 31 000 and 31 500, suggesting that the enzyme was active as a heterodimer. Two-dimensional electrophoresis gave four protein spots which were subjected to tryptic digestion. Sequence analysis of the resulting peptides by ESI-MS/MS revealed that all four polypeptides were isoforms of the elongation factor 2, no sequence similarity was observed with prenyltransferases.Hyperforin ist ein Breitband-Neurotransmitter-Wiederaufnahmehemmer aus Hypericum perforatum (Johanniskraut). Unter Benutzung von DEAE- und Mono-QAnionenaustauschchromatographie, Hydroxylapatit -Adsorptionschromatographie, Hydrophober Interaktionschromatographie und Gelfiltration wurde das Enzym gereinigt. Auf einer kalibrierten Gelfiltrationssäule ergab sich für die Dimethylallyltransferase eine relative Molekularmasse von 63000. In SDS-Polyacrylamidgel ergaben sich zwei benachbarte Polypeptidbanden mit Mr 31000 und 31500, was vermuten ließ, dass das Enzym als Heterodimer aktiv ist. Das gereinigte Protein trennte sich, einer 2-D- Elektrophorese unterworfen, in vier Proteinspots, die tryptisch verdaut werden. Die Sequenzanalyse der erhaltenen Peptide ergab, dass alle vier Polypeptide Isoformen des Elongationsfaktors 2 darstellen. Eine Ähnlichkeit zu Prenyltransferasen wurde nicht festgestellt
