Ortsgerichtete Mutagenese von Hypericum Benzophenon-Synthasen

Benzophenone derivatives are important class of phenolic natural products with complex chemical structures and important pharmacological activities. Their C13 skeleton formed from one molecule of benzoyl-CoA and three molecules of malonyl-CoA. This reaction is catalyzed by benzophenone synthase (BPS), which is a member of type III polyketide syntheses. Recently, H. androsaemum BPS has been transformed into phenylpyrone synthase. Several BPSs were cloned from different Hypericum species such as H. sampsonii, H. calycinum and H. perforatum ssp angustifolium. Interestingly, all of the cloned Hypericum BPSs maintain the same amino acids lining the active site cavities especially Thr135.Various site-directed mutagenesis approaches were applied to replace Thr135 in H. sampsonii BPS (HsBPS) for the other 19 amino acids. A single amino acid substitution in the active site cavity (HsT135K) transformed H.sampsonii BPS into 4-hydroxycoumarin synthase which accepts salicoyl-CoA forming 4-hydroxycoumarin This is a novel PKS function and further expands the biosynthetic repertoire of the PKSs super family. An updated homology model was constructed as a dimer based on the crystal structure of M.sativa CHS2 to rationalize that dramatic functional variation of HsT135K mutant. The modeling showed that, lysine residue has a long charged side chain which is in contact with the adjoining monomer and the dimer interface. Lysine in position 135 does not only acts as an active site residue but also extended to the other monomer and the dimer interface Therefore, X-ray crystal structure is required for the wild and the mutant to explain this dramatic change in details. Replacement of Thr135 with Lys in H. calycinum, H. perforatum ssp angustifolium and H. androsaemum BPSs resulted in, no over-expression in case of HpaT135K, no soluble protein in case of HcT135K. However, generated HCS with HaT135K but with much lower activity compared to the HsT135K mutant. Leucine and isoleucine in the same position convert H. sampsonii, calycinum and perforatum ssp angustifolium into functional PPSs. Der Austausch von Thr135 gegen Lysin in den H. calycinum, H. perforatum ssp angustifolium and H. androsaemum BPSs resultierte in keiner Überexpression von HpaT135K und keinem löslichen Protein für HcT135K. Jedoch erwies sich HaT135K als eine HCS, aber mit geringerer katalytischer Aktivität als die HsT135K-Mutante.
