Higher electrophoretic mobility band of pUC19 (hmP19) DNA is not double-stranded in nature. pUC19 plasmid was denatured using 0.5 N NaOH, pH was neutralized using 0.5 M HCl, and DNA was incubated with Hind III, which degrades double-stranded DNA. Native pUC19, but not hmP19 DNA was degraded by Hind III (lane 7). Experiment was repeated three times (a, b, c). Lane 1, 1 kb ladder; Lane 2, pUC19; Lane 3, pUC19 + 0.5 N NaOH; Lane 4, pUC19 + 0.5 N NaOH + 0.5 M HCl; Lane 5, pUC19 + Hind III; Lane 6, pUC19 + 0.5 N NaOH + Hind III; Lane 7, pUC19+ 0.5 N NaOH + 0.5 M HCl + Hind III. </p
