Addition of sodium hydroxide (NaOH) to pBR322 plasmid resulted in the distinct band of higher electrophoretic mobility DNA. Approx. 5 µg plasmid DNA was added with equal volume (5 µl) of NaOH solution of increasing concentration. After 4-5 min, DNA was analyzed by agarose gel electrophoresis. A distinct higher electrophoretic mobility band was observed in the plasmid DNA added with 0.5 N NaOH (marked by arrow, lane 5). Experiment was repeated at least two times (a, b). Lane 1, Ladder; Lane 2, pBR322 plasmid; Lane 3, pBR322 plasmid + 0.10 N NaOH ; Lane 4, pBR322 plasmid + 0.25. N NaOH; Lane 5, pBR322 plasmid + 0.50 N NaOH. </p
