Formulation of sustained-release aspirin capsules

Asetýlsalisýl sýra eða aspirín er NSAID lyf sem hefur verið lengi notað í lækningaskyni. Asetýlsalisýl sýra hefur bólgueyðandi, blóðþynnandi og hitalækkandi áhrif með því að hindra cyclooxygenasa viðtaka líkamans, ósértækt. Markmið rannsóknarinnar var að hanna lyfjaform með asetýlsalisýl sýru sem virka efnið með stýrðri losun í fjórar klukkustundir. Markmiðið var að asetýlsalisýl sýra átti að vera 45-55% losuð eftir 45 mín, 65-75% eftir 120 mín og 90-100% eftir 240 mín. Stýrð losun er notuð til þess að fá ákveðinn styrk af lyfi í líkamanum á ákveðnum tíma. Stýrð losun getur verið ákjósanleg þegar markmið lyfjagjafar er að lengja viðverutíma lyfs í líkamanum. Til þess að ná fram þessum eiginleikum voru framleidd hylki, með nokkurs konar forðaverkun. Forðaverkunnni var náð með því að nota gelatín hylki og fjölliðu sem að bólgnar út í viðveru vatns. Gelatín hylkið leysist upp og skilur eftir sig fjölliðu lagað hylki. Þar sem að asetýlsalisýl sýra er óleysanleg í vatni þá leysist það frá fjölliðunni háð niðurbroti hennar. Tuttugu mismunandi lyfjaform voru prófuð, þar af sextán töflur og fjögur hylki. Í lokin var besta lyfjaformið valið og prófanir framkvæmdar samkvæmt Evrópsku lyfjaskránni og stóðust hylkin allar þær prófanir.Acetylsalicylic acid, or aspirin, is an NSAID drug that has been used in medicine for a long period of time. Acetylsalicylic acid has anti-inflammatory properties, anti-platelet effects and fever-reducing properties by non-selectively inhibiting the cyclooxygenase receptors of the body. The main goal of this project was to produce a formulation containing acetylsalicylic acid, asetylsalicylic acid, with a sustained-release of four hours. The active ingredient, acetylsalicylic acid, should be 45-55% released by 45 minutes, 65-75% by 120 minutes, and 90-100% by 240 minutes. Sustained-release formulations can be used to achieve a specific concentration of the drug in the body at a certain time. A sustained-release dissolution profile can be optimal if the goal is to prolong the presence of the drug in the body. A capsule formulation was used to achieve sustained-release with depot properties. The depot properties were achieved by using gelatine capsules and a water-swellable polymer. The gelatin capsule dissolves and leaves a capsule-shaped polymer matrice. Acetylsalicylic acid is insoluble in water, therefore its dissolution from the polymer is dependent on its decomposition. Twenty formulations were made, sixteen tablet formulations, and four capsule formulations. A decision was made to move forward using capsules for this project because they gave more promising results. All nessecery tests were performed on the final formulation according to the standards of the European Pharmacopoeia and all capsules passed those tests
