Synthesis, isolation and physicochemical characterization of complex compounds between O-, N-organic sbstrates and metallotoxins

The aim of this study is the synthesis, isolation and physicochemical characterization of complex compounds between O-, N- organic substrates and metallotoxins. More specifically, this work-project investigates the ability of the toxic metal ions Pb(II) and Cr(III) to a) promote chemistry with O-, N-containing physiological and physiologically relevant organic substrates in aqueous solution, and b) pursue the physicochemical characterization of new binary and ternary species in the solid state and in solution. The ultimate scope of the work is to shine light upon the nature of interactions developing between the toxic metal ions (Pb(II) and Cr(III) and the organic substrates, in view of their potential molecular toxicity in a biological setting and their potential to serve as synthons in the synthesis of advanced materials. The first part of the thesis describes the study of the new binary and ternary arisen compounds of the toxic metal ion Pb(II). A plethora of select organic substrates were employed. Most of them are low-molecular-mass physiological ligands, including succinic acid, malic acid, citric acid, fumaric acid, etc. (they are components of the Krebs cycle). The early results of this study led to the general idea of using more complex organic ligands in order to synthesize and isolate novel compounds with a) a variety of structural architectures and topologies, and b) very interesting physicochemical properties. In fact, the herein obtained results were unexpected and often intriguingly more interesting than the ones expected. Another remark, worth pointing out, is the employment of a variety of bases for the pH adjustment. When the bases used were organic, such as 1,10-phenanthroline, the results were very impressive. As a matter of fact, unique binary and ternary materials were synthesized and isolated, with 1,10-phenanthroline being coordinated to the metal ion and bestowing interesting structural and spectroscopic (fluorescence) properties. Equally remarkable is the fact that the arisen coordination polymers constitute one of the most important classes of inorganic-organic hybrid materials, which have attracted considerable research attention not only because of their intriguing variety of architectures and topologies but also because of their fascinating potential applications in functional solid materials, ion exchange, catalysis, and the development of optical, electronic, and magnetic devices. Therefore, good knowledge of Pb(II) coordination properties toward select organic substrates ligands, including the lone pair of electrons, the diverse coordination number and coordination geometry, are crucial factors in understanding the toxicological properties of Pb(II) at the molecular level. In contrast to transition metals, the main group Pb(II) ion possesses a unique coordination preference toward variable nature substrates rarely observed in the rest of the periodic table, thereby presenting unique opportunities for the design and development of novel structures with new, unique and interesting characteristics. Collectively, the aforementioned reflect a wide research interest in the investigation on Pb(II) chemistry toward physiological or physiologically relevant substrates not only because it is a very toxic metal and very harmful to human health, but also because well-defined Pb(II) complexes are very important in the development of the science and technology of advanced materials. In the second part of this research work, the ability of Cr(III) to interact with organic substrates, such as citric acid, quinic acid, and 2-hydroxyethyliminodacetic acid was investigated in depth. The outcome of these efforts was the isolation of many new compounds including: (NH4)4[Cr(C6H4O7)(C6H5O7)].3H2O, Na3[Cr(C6H5O7)2]. 8.5H2O, (NeoΗ)6[Cr(C6H5O7)2](ΝΟ3)3 .14H2O, Na[Cr3O(quinic)6(H2O)3](NO3)1.5(OH)0.5 .6H2O, (NH4)[Cr{HOCH2CH2N(CH2COO)2}2].2H2O, Na[Cr{HOCH2CH2N(CH2COO)2}2].H2O, (NeoΗ)2[Cr{HOCH2CH2N(CH2COO)2}2].(OH)(H2O) and [Cr(C6H9NO5)(phen)(H2O)](NO3).3H2O. All of the new species were further characterized by elemental analysis, spectroscopic, structural, thermal and magnetic susceptibility studies. For the binary systems of Cr(III)-citrate and Cr(III)-quinic potentiometric studies and aqueous speciation studies were carried out in order to clarify and delineate the different species emerging from the interactions of Cr(III) with the two hydroxycarboxylic acids in aqueous media. Aqueous speciation (at the binary or ternary level) provides a clear view of the distribution of species present in the entire pH range investigated and gives the opportunity to develop strategic approaches for the synthesis, isolation and physicochemical characterization of the participating species as function of pH and molecular stoichiometry of the interacting reagents. The collective approach reflects the quintessence of structural speciation. All of the new isolated compounds of Cr(III) are soluble in water. The variety of applications of chromium and its effects on human health, necessitate in-depth knowledge of soluble and bioavailable species of Cr(III), as a result of the binary and ternary interactions with proper physiological substrates, and the determination of the physicochemical properties of the so arisen species as a function of pH and molecular stoichiometry. Bearing in mind the aforementioned facts, the aqueous chemistry of chromium, in its most dominant oxidation states Cr(VI) and Cr(III), can be a challenging research area to probe and a complex one to associate with biological and non-biological processes. Finally, the simplicity of the nature of the herein isolated novel species of Cr(III) proclaims strongly the employment of such species, as staring materials for the rational design and synthesis of diversely composed advanced materials bearing specific physicochemical properties justifying technological applications.Η παρούσα διδακτορική διατριβή πραγματεύεται τη σύνθεση, απομόνωση και τον φυσικοχημικό χαρακτηρισμό ενώσεων μεταξύ Ο-, Ν-οργανικών υποστρωμάτων και μεταλλοτοξινών. Πιο συγκεκριμένα, μελετάται η δυνατότητα των τοξικών μεταλλοϊόντων του μολύβδου (Pb(II)) και του χρωμίου (Cr(III)) σε υδατικά διαλύματα να αλληλεπιδράσουν με Ο-, Ν-οργανικά υποστρώματα. Δίδεται έμφαση στην φύση των αλληλεπιδράσεων μεταξύ των τοξικών μεταλλοίόντων (Pb(II) και Cr(III)) και των οργανικών υποστρωμάτων (substrates). Το πρώτο μέρος της διδακτορικής διατριβής πραγματεύεται τη μελέτη νέων ενώσεων του τοξικού μεταλλοϊόντος Pb(II). Στην μελέτη αυτή χρησιμοποιήθηκαν πληθώρα οργανικών υποστρωμάτων. Τα περισσότερα από αυτά ανήκουν στην ομάδα των φυσιολογικών υποστρωμάτων, όπως είναι το ηλεκτρικό, το μηλικό, το κιτρικό, το φουμαρικό οξύ, κ.ά. (συμμετέχουν στον κύκλο του κιτρικού οξέος – κύκλο του Krebs). Ωστόσο, τα πρώτα αποτελέσματα της μελέτης αυτής οδήγησαν στη γενική σκέψη χρησιμοποίησης περισσότερο πολύπλοκων οργανικών υποστρωμάτων για την απομόνωση σύνθετων υλικών από πλευράς αρχιτεκτονικής και τοπολογίας δομών και φυσικά με πολύ πιο ενδιαφέρουσες ιδιότητες. Πράγματι, τα αποτελέσματα ήταν τα αναμενόμενα και σε πολλές περιπτώσεις πολύ πιο ενδιαφέροντα. Ένα άλλο σημείο, το οποίο πρέπει να επισημανθεί για τη μελέτη των ενώσεων του μολύβδου, είναι η χρήση διαφόρων βάσεων για την αύξηση του pH του διαλύματος. Όταν οι βάσεις που χρησιμοποιήθηκαν ήταν κάποια οργανικά μόρια, όπως αυτό της 1,10- φαινανθρολίνης, τα αποτελέσματα όσον αφορά τις δομές που προέκυψαν ήταν αδιαμφισβήτητα εντυπωσιακά. Η ερευνητική αυτή δραστηριότητα οδήγησε στην απομόνωση μοναδικών υλικών, στα οποία το μόριο της 1,10-φαινανθρολίνης συναρμόστηκε και προσέδωσε σε αυτά ιδιαίτερες ιδιότητες, όπως είναι αυτή του φθορισμού. Αξιοσημείωτο είναι το γεγονός ότι τα περισσότερα υλικά που έχουν απομονωθεί είναι πολυμερικά. Τα συναρμοσμένα πολυμερή αποτελούν μια από τις πιο σημαντικές κατηγορίες οργανικών-ανόργανων υβριδικών υλικών, τα οποία έχουν κεντρίσει μεγάλο ερευνητικό ενδιαφέρον όχι μόνο για τις διαφορετικές αξιοθαύμαστες αρχιτεκτονικές και τοπολογίες που αναπτύσσουν, αλλά και για τις συναρπαστικές πιθανές τους εφαρμογές σε λειτουργικά στερεά υλικά, σε ιοντοανταλλαγές, στην κατάλυση και στην ανάπτυξη οπτικών, ηλεκτρονικών και μαγνητικών συσκευών. Όλα αυτά προκαλούν το ερευνητικό ενδιαφέρον να ασχοληθεί κανείς με το μεταλλοϊόν του Pb(II), όχι μόνο γιατί είναι τοξικό και ιδιαίτερα επιβλαβές για την υγεία, αλλά και γιατί οι ενώσεις του είναι ιδιαίτερα σημαντικές στην περαιτέρω εφαρμογή τους στην επιστήμη και τεχνολογία υλικών. Στο δεύτερο μέρος της διδακτορικής διατριβής μελετάται η δυνατότητα αλληλεπίδρασης του Cr(III) με οργανικά υποστρώματα, όπως το κιτρικό οξύ, το κινικό οξύ και το 2-υδροξυ- αιθυλο-ιμινοδιοξικό οξύ. Αποτέλεσμα της αλληλεπίδρασης αυτής είναι η απομόνωση πολλών νέων ενώσεων, μεταξύ των οποίων συμπεριλαμβάνονται οι εξής: (NH4)4[Cr(C6H4O7)(C6H5O7)].3H2O, Na3[Cr(C6H5O7)2].8H2O, (NeoΗ)6[Cr(C6H5O7)2](ΝΟ3)3 .14H2O, Na[Cr3O(quinic)6(H2O)3](NO3)1.5(OH)0.5 .6H2O, (NH4)[Cr{HOCH2CH2N(CH2COO)2}2].2H2O, Na[Cr{HOCH2CH2N(CH2COO)2}2].H2O, (NeoΗ)2[Cr{HOCH2CH2N(CH2COO)2}2].(OH)(H2O), και το [Cr(C6H9NO5)(phen)(H2O)](NO3).3H2O. Ακολούθησε δομικός χαρακτηρισμός με κρυσταλλογραφία ακτίνων Χ, φασματοσκοπική μελέτη με φασματοσκοπία ηλεκτρονικού παραμαγνητισμού συντονισμού (EPR), φασματοσκοπία υπερύθρου και ορατού/υπεριώδους, στοιχειακή ανάλυση, θερμοσταθμική ανάλυση, ενώ πραγματοποιήθηκαν μελέτες μαγνητικής επιδεκτικότητας και κυκλικής βολταμμετρίας για όλες τις παραπάνω ενώσεις. Για τα συστήματα του Cr(III)-κιτρικό και Cr(III)-κινικό έχουν γίνει ποτενσιομετρικές μελέτες ως μέρος των πειραμάτων για την εξιχνίαση της υδατικής ειδογένεσης (aqueous speciation), με σκοπό τη μελέτη και κατανόηση των εν δυνάμει ειδών που προκύπτουν από την αλληλεπίδραση του Cr(III) με τα δύο αυτά οξέα στο υδατικό διάλυμα. Η υδατική ειδογένεση προσφέρει πλήρη εικόνα για τα είδη που υπάρχουν σε όλο το εύρος του pH και δίνει την ευκαιρία ανάπτυξης συνθετικών στρατηγικών για τη σύνθεση, απομόνωση και φυσικοχημικό χαρακτηρισμό των συμμετεχόντων ειδών. Η συλλογική αυτή τακτική αποτελεί την πεμπτουσία της δομικής ειδογένεσης (structural speciation). Όλα τα υλικά του χρωμίου που προέκυψαν είναι διαλυτά στο νερό. Η ποικιλία των εφαρμογών του Cr και οι επιδράσεις που έχει αυτό στην υγεία, απαιτούν τη γνώση της φύσης των διαλυτών και βιοδιαθέσιμων ειδών του Cr που αναπτύσσονται ως αποτέλεσμα δυαδικών και τριαδικών αλληλεπιδράσεων με τα απαραίτητα υποστρώματα, καθώς επίσης και τον προσδιορισμό των φυσικοχημικών ιδιοτήτων των ειδών αυτών ως συνάρτηση του pH. Mε γνώμονα τη διαπίστωση αυτή, γίνεται η σύνδεση της υδατικής χημείας του χρωμίου, στις επικρατέστερες οξειδωτικές καταστάσεις, με βιολογικές και μη βιολογικές διεργασίες. Επιπλέον, η απλότητα της φύσης των υλικών του χρωμίου που αναδύθηκαν, καθιστά τη χρήση των ειδών αυτών, ως αρχικών υλικών, πολύ ικανή για τη σύνθεση νέων προηγμένων υλικών με ιδιαίτερες ιδιότητες για επιλεγμένες τεχνολογικές εφαρμογές
