Despite compelling evidence that the phenotypic modulation of vascular smooth muscle (VSM) cells plays a key role in the development and progression of arteriosclerosis, we are just beginning to unravel the molecular mechanisms and factors that control this process. It is proposed that the duration of an epidermal growth factor receptor (EGFR)-mediated ERK1/2 (extracellular signal- regulated kinases 1/2) phosphorylation controls the phenotypic outcome of a cell. In that context, long-lasting ERK1/2 kinetics, as opposed to short-lived kinetics, have been linked to cellular differentiation in various cell types.
