<p>Jurkat cells were transfected with 1 µg GFP in pCDNA3.1/Zeo+ and either 6 µg NS3/4A/4B expression plasmid (red line) or CAT expression plasmid (blue line) and scrambled anti-CD4 siRNA. Equal numbers of viable cells were labeled with anti-CD4 PE-Cy7 and anti-CXCR4-APC and analyzed by FACS on days 3 (<b>A</b> and <b>B</b>), 4 (<b>C</b> and <b>D</b>), and 5 (<b>E</b> and <b>F</b>) post transfection. Cells were gated for GFP expression and then analyzed for CD4 and CXCR4 expression. As a control to demonstrate reduction in receptor expression, cells were transfected with 1 µg GFP in pCDNA3.1/Zeo+ and 6 µg CAT expression plasmid plus anti-CD4 siRNA (<b>A,C,E</b>: gray shaded area, <b>B,D,F</b>: black line).</p
