DNA vaccination against classical swine fever virus-influence of immune stimulators on the vaccine efficacy

Das Hauptziel dieser Arbeit, bestand in der Herstellung einer wirksamen CSFV-spezifischen DNA-Vakzine. Hierfür wurde das E2-Glykoprotein des CSFV in eine Reihe verschiedener eukaryontischer Expressionsvektoren kloniert die sich in der Auswahl der verwendeten Promotoren (CMV oder RSV) sowie in den zusätzlich inserierten Genen von Zytokinen und immunstimulatorischen Molekülen (IL-12, IL-18, CD40L) unterschieden. Die anschließende Überprüfung der Expression der E2, IL-12, IL-18 und CD40L-Gene, in den Konstrukten, erfolgte mittels Transfektion in Zellen und anschließender Immunfluoreszenzanalyse bzw. durch funktionelle Tests. In den Vakzinierungsversuchen im Schwein wurde in einem ersten Schritt die Applikationsart sowie die Infektionsdosis optimiert. Danach wurde die Schutzwirkung der E2-Grundkonstrukte im im Verhältnis zur Schutzwirkung von E2-Konstrukte die zusätzlich die Sequenz eines immunstimulatorischen Moleküls enthielten analysiert.Ein weiterer Aspekt dieser Arbeit bestand in der pro- und eukaryontischen Expression der für die DNA-Vakzine eingesetzten immunstimulatorischen bzw. CSFV-spezifischen Moleküle. Im Verlauf dieses Projektes gelang es, porcines IL-18, den CD40L sowie die p40 Untereinheiten des IL-12, prokayontisch zu exprimieren, aufzureinigen und über Western Blot bzw. Immunfluoreszenz nachzuweisen. Das exprimierte und aufgereinigte IL-18 und der CD40L wurden zur Gewinnung spezifischer polyklonaler Seren eingesetzt. Für die p35-Untereinheit des IL-12-, und das E2-Glykoprotein gelang ein Nachweis der Expression als GST-(p35) bzw. (E2)-Fusionsprotein durch Western Blot Analyse. Außerdem wurden für die eukaryontische Expression der Moleküle stabile 293-Zelllinien hergestellt und die Expression der CD40L, IL-18 und IL-12-Gene durch Immunfluoreszenzanalyse, Radioimmunpräzipitation, Western Blot bzw. über funktionelle Tests nachgewiesen.The aim of this study was the construction of an effective CSFV specific DNA-vaccine. For this purpose, the E2-glycoprotein gene of the CSFV was cloned into a number of different eukaryotic expression vectors which vary in the selection of the used promotors (CMV or RSV) as well as in the additionally inserted genes of cytokine and immunstimulatory molecules. The expression of the E2, IL-12, IL-18 and CD40L-gene, in the constructs, were examined after transfection in cells by immune fluorescence analysis or functional tests. The first experiments of this work based on the optimization of the route of vaccination as well as the infection dose in pigs. Afterwards the protective effect of the E2 constructs in comparison to the protective effect of E2 constructs which additionally contained sequences of a immunstimulatory molecule (IL-12, IL-18, CD40L) were analysed. A further aspect of this work was the eukaryotic and prokaryotic expression of the immunstimulatory or CSFV specific molecules used for the DNA vaccination. In this study the porcine IL-18, the CD40L as well as the p40 subunits of the IL-12 were prokaryotic expressed and tested by Western Blot or immunfluorescenc analysis. The expressed and purified IL-18 and the CD40L were used for the production of a specific polyclonal serum. The p35 subunit of the IL-12 -, and the E2-glycoprotein were expressed as a GST (p35) or (E2)-fusion protein and the gene expression were confirmed by Western Blot analysis. For the eukaryotic expression of the molecules, stable 293-cell line was prepared and the expression of the CD40L, IL-18 and IL-12-gene was verified by immunefluorescence analysis, radioimmun precipitation, Western Blot or by functional tests
