Identification of B- and T-cell epitopes of classical swine fever virus with synthetic peptides

Die klassische Schweinepest ist eine virale Erkrankung, die unter natürlichen Bedingungen nur beim Haus- und Wildschwein auftritt. Der Erreger der klassischen Schweinepest ist das 'classical swine fever virus' (CSFV), das eine der ökonomisch bedeutendsten Tierseuchen verursacht. Die Identifizierung von B- und T-Zell-Epitope des Klassischen Schweinepestvirus ist eine Voraussetzung für die Entwicklung einer Peptidvakzine. Im Rahmen dieser Arbeit wurden B- und T-Zell-Epitope des Klassischen Schweinepestvirus (CSFV) mit Hilfe von synthetischen Peptiden im natürlichen Wirtstier identifiziert. 573 Pentadekapeptide, die ca. 80 des kodierenden Genombereiches von CSFV-Stamm Glentorf abdecken, wurden mit einer Überlappung von 10 Aminosäuren synthetisiert. Ein Schwerpunkt war die Etablierung eines Peptid-ELISA mit dem lineare B-Zell-Epitope nachgewiesen werden konnten. Um diese B-Zell-Epitopkartierung durchführen zu können, wurden die Pentadekapeptide im ELISA eingesetzt. Ein lineares B-Zell-Epitop wurde auf dem Autoprotease Protein des CSFV identifiziert. In Teil 2 der Arbeit gelang der Nachweis von T-Zell-Epitopen mit Hilfe der überlappenden Pentadekapeptide. 26 Peptide konnten mit peripheren mononukleären Blutzellen (PBMC) eines mit dem CSFV-Stamm Glentorf infizierten Schweines als positiv identifiziert werden. 18 von diesen 26 Peptiden wurden von PBMC eines mit dem CSFV-Stamm Alfort/187 infizierten Schweines in Proliferationsassays ebenfalls erkannt. Das Peptid 290 aus dem NS2-3 Bereich des CSFV, das in den Proliferationsassays den höchsten Stimulationsindex zeigte, wurde weiter untersucht. Wie mit Hilfe von ELISPOT-Tests gezeigt werden konnte, war das Peptid 290 in der Lage CSFV-sensibilisierte T-Zellen zur IFN-?-Produktion anzuregen. Blockierungsexperimente mit monoklonalen Antikörpern gegen an der Antigenpräsentation beteiligte Moleküle (CD4, CD8, MHC-I, MHC-II), zeigten eine Aktivierung sowohl der T-Helfer-Zellen als auch der CTL nach Stimulation mit diesem PeptiCSFV (classical swine fever virus) causes a fatal disease of swine and its outbreak has severe economic consequences. Identification of B- and T-cell epitopes on the viral proteins is necessary for the development of CSFV peptide vaccines. In order to identify B- and T-cell epitopes of CSFV, 573 pentadecapeptides covering about 80 of viral protein sequences strain Glentorf and overlapping each other by 10 residues were synthesised and the immune response of the natural host to these peptides was monitored. An important technical aspect of this work was the development of a peptide-ELISA that can be used for identification of linear B-cell epitopes. Using this assay, the pentadecapeptides were screened for the presence of linear B-cell epitopes and one such epitope on the autoprotease of CSFV was identified. In the second part of this work, these peptides were screened for the identification of T-cell epitopes. Twenty-six peptides were identified as positive in proliferation assays with peripheral blood mononuclear cells (PBMC) from a CSFV-Glentorf infected d/d haplotype pig. Eighteen out of these 26 peptides were also recognized by PBMC from a CSFV-Alfort/187 infected d/d haplotype pig, suggesting that these peptides are immunogenic in both CSFV infected animals. The peptide 290 derived from NS2-3 region of CSFV showed the highest stimulation index in the proliferation assays. The ability of this peptide to induce IFN-? secretion and its MHC restriction of proliferative response were further characterised. This peptide induced IFN-? secretion after secondary in vitro restimulation as monitored by ELISPOT-assays. To analyse the MHC restriction of the proliferative response to peptide 290, monoclonal antibodies against CD4, CD8, MHC-I and MHC-II molecules were used.
