Zahlreiche klinische Studien zeigen die positiven Effekte der Bisphosphonate auf metabolische und tumorassoziierte Knochenkrankheiten. Das erste Bisphosphonat, Etidronat, ist seit 30 Jahren bekannt. Seitdem wurden viele neue Bisphosphonate mit größerem Wirkungspotential entwickelt. Deren Wirkungsmechanismus auf Osteoklasten ist vielfach untersucht worden und hinreichend bekannt. Ihre Einwirkung auf Osteoblasten hingegen ist bisher größtenteils unbekannt.
