Development of Marine-derived Protein-based Nanoparticles for Photothermal Cancer Therapy

MasterCancer has been a leading cause of human death worldwide, and various therapeutic therapies have been studied and developed. Surgery, radiation, and chemotherapy have traditionally been used as three standard anticancer therapies, however there is a limit to complete removal of the cancer by surgery, and anticancer drugs may cause side effects on normal cells. Drug resistant cells make it more difficult to treat cancer by chemotherapy. In addition, the poor quality of life is the main reason for patients to hesitate to take radiation therapy. Accordingly, new anti-cancer therapies have been developed with little concern about side effects and complications. Especially, photothermal cancer treatments that can kill cancer cells through light-induced hyperthermia have been attracted considerable attention due to high selectivity, and minimally invasiveness. Using photothermal agents, which can convert light to thermal energy, heat, can be generated locally, increasing the temperature of cancer cells above 42.5 ℃ to induce apoptosis. Additionally, combined treatments with chemotherapy and photothermal therapy have been reported to improve therapeutic efficacy synergistically through different mechanism of action. Inorganic materials having high photothermal conversion efficiency such as gold nanoparticles (NPs) and iron oxide NPs are widely used as photothermal therapeutic agents, but there are limitations in actual clinical applications due to their toxicity. Therefore, research has been developed for finding small molecules having high photosensitivity and biocompatibility. Indocyanine green (ICG), a FDA-approved safe substance, has a photosensitizing property with near infrared (NIR) light that has high skin permeability. However, rapid degradation of aqueous media (short half-life), low thermal stability, and its easy binding to blood proteins are the limitations in practical applications. In order to improve the stability of ICG, many researches are trying to deliver ICG using NPs. However, there are problems with immune response of NPs as well as the matter of toxicity come from high doses. Therefore, in order to further highlight the advantages of biocompatible ICG, there is a need to have high biocompatibility of the nanocarrier itself. In this study, we encapsulated ICG in highly stable NPs that are made of marine organism-derived proteins with dityrosine binding. The ultimate goal of this study was to maximize the photothermal effect of ICG and at the same time, minimize the side effects with high biocompatible protein-based nanoparticles. In detail, mussel adhesive protein (MAP) and sea anemone silk-like protein (aneroin) were used with different delivery system according to the characteristics of two materials. First, MAP-based NPs (MNPs) produced by electrospray through enzyme-catalyzed dityrosine crosslinking and confirmed to have a uniform size with excellent adhesive property. Cytotoxicity experiments also demonstrated the biocompatibility of the MNPs. The ICG was encapsulated in MNPs with high efficiency, and the ICG-loaded MNPs (M@ICG NPs) had a higher temperature rise rate than free ICG and demonstrated successful anticancer effect on breast cancer cells, MCF7. The unique adhesion property of MAP can be applied to the post-operative site or by intratumoral injection to maintain high concentrations of photothermal agents at the local site for effective anticancer treatment effect. Next, we developed aneroin-based NPs (ANPs) by electrospray combining dityrosine crosslinking through photo-oxidative reaction, we successfully fabricated ANPs with uniform particle size distribution, and verified low cytotoxicity, and passive tumor targeting property of ANPs via the enhanced permeability and retention (EPR) effect that NPs tend to accumulate in tumor much more than in normal tissues. These results confirmed the possibility of using it as a photothermal agent that can be administered systemically by intravenous injection. Overall, we have developed the two types of marine organism-derived proteins-based NP systems for photothermal therapy to maximize anticancer treatment efficacy with minimal side effects. Both bioengineered proteins have advantages of excellent biocompatibility, biodegradability, and mass production. In addition, molecular biotechnology also makes it possible to produce homogeneous proteins and add functional peptides easily. The NPs developed in this dissertation showed the possibility to be applied successfully to photothermal therapy by protecting ICG through stable dityrosine binding for a long time without releasing. In addition, these NPs are anticipated to be used throughout local or systemic cancer therapies not only because of their superior biocompatibility and biodegradability, but also due to their unique underwater adhesion property of MAP and the excellent physical property of aneroin.전세계적으로 암은 많은 사람들을 고통받게 하는 질병으로 꾸준히 인류의 주된 사망 원인이었다. 3대 표준 항암 치료 요법인 수술, 방사선 조사, 그리고 화학 요법이 전통적으로 활용되고 있지만, 수술로는 미세한 암세포의 제거가 어려워 완전한 암 치료에 한계가 있으며, 항암제의 경우 정상적인 세포에도 부작용을 일으킬 수 있으며, 약물내성세포의 경우 치료가 더 어렵다. 또한, 삶의 질 저하를 야기하는 방사선 치료 부작용에 대해 우려하는 환자에게는 이 치료법들만을 적용하기 어려운 것이 현실이다. 이에 따라 항암치료 부작용과 합병증에 대한 우려가 적은 새로운 항암 치료법들이 개발되어왔고, 그 중에서도 레이저를 통하여 암 병변에 국소적으로 42.5도가 넘는 열을 발생시켜 열에 민감한 암세포만 선택적으로 파괴하는 광열암치료가 해결책 중 하나로 주목받고 있다. 광열암치료는 빛을 조사하여 발생한 열로 암세포가 스스로 괴사하도록 유도하기 때문에 면역력을 높이고, 단독 치료만으로도 효과를 볼 수 있지만, 화학 요법과 병행될 경우 항암제가 종양 세포에 더욱 효과적으로 도달할 수 있도록 기여하며 상승효과를 줄 수 있다. 광열 치료제로써 높은 광열전환효율을 가진 금 나노입자, 산화철 나노입자 등 무기물질이 많이 이용되고 있지만 독성의 위험성으로 인하여 실제 임상 적용에는 한계가 있다. 이에 생체적합성을 지닌 유기물 기반의 저분자 물질에 대한 관심이 높아지고 있으며, 그 중에서도 특히 Indocyanine green(ICG)는 FDA 승인을 받은 물질로 높은 생체적합성을 가지고 있다. 오래전부터 의료계에서 조영제로 사용되어온 만큼 안전성이 확보되어 있으며 피부 투과성이 높은 근적외선 빛에서 광열효과를 가지고 있기 때문에 생체적합성이 높은 광열제로서 높은 가능성을 지니고 있다. 하지만 수중 매질에서의 낮은 안정성과 빠른 세포 흡수, 그리고 짧은 반감기로 인해 실제 응용에는 한계점이 있다. 이에 ICG의 안정성을 높이기 위해 나노입자를 이용해 전달하고자 하는 연구들이 많이 이루어져왔지만, 나노 입자 자체 혹은 가교제의 독성 및 면역 반응에 대한 문제와 광열 치료제 자체의 열적 한계로 인한 불충분한 발열량과 높은 투여량에 대한 문제들을 가지고 있기도 하다. 따라서 생체적합성을 지닌 ICG의 장점을 더욱 부각시키기 위해서는 이를 전달하는 나노캐리어 또한 높은 생체적합성을 지닐 필요성이 있으므로 단백질 기반의 나노 입자에 탑재하는 연구가 필요하다. 단백질 기반의 나노 입자는, 우선 단백질이라는 재료로부터 높은 생체적합성과 생분해성을 지니며, 다양한 작용기를 가지고 있어 여러 반응들을 통하여 생체재료 혹은 생체 의료용 소재로 제작하기 용이하다. 또한 분자생물공학을 활용하여 재생산된 단백질을 활용할 경우, 독특한 성질을 지닌 단백질을 균일한 상태로 대량생산할 수 있다는 장점이 있으며, peptide를 fusion시킴으로써 추가적인 기능도 부여할 수 있다. 결론적으로, 본 연구는 광열 효과가 높으면서도 FDA 승인을 받은 안전한 물질이지만, 수성 매질의 빠른 분해 및 신체로부터의 빠른 제거 및 혈액 단백질에 쉽게 결합하기 때문에 실질적인 활용에 제한이 있는 ICG를 다이타이로신 결합 기반의 매우 안정적인 단백질 나노입자에 탑재함으로써 ICG의 광열 치료 효과를 극대화하고, 생체적합성이 높은 단백질 기반의 나노입자로서 부작용을 최소화하는 것을 목표로 진행되었다. 본 연구에서는 자체 기술을 통하여 생산한 두 가지 단백질, 홍합접착단백질(MAP)과 아네로인을 활용하여 연구를 진행하였다. 홍합 접착 단백질 기반 나노입자는 효소에 의해 다이타이로신 결합을 형성하며 균일한 크기를 가짐을 확인하였으며, 접착력 역시 우수함을 확인하였다. 세포 독성 실험을 통하여 홍합 접착 단백질 기반 나노입자 자체의 생체 적합성 역시 증명하였다. ICG를 coelectrospray 기법을 통하여 다양한 몰비율로 탑재한 결과, MAP와 ICG의 몰 비율이 1:6이 될 때까지도 95%가 넘는 높은 효율로 ICG를 탑재함을 확인하였다. In vitro 실험결과, 탑재되지 않은 ICG 보다 더욱 빠른 온도 상승 속도를 보이며 유방암 세포주인 MCF7에서 성공적인 항암 효능을 보였다. 따라서, 홍합접착단백질 기반의 나노입자는 홍합 접착 단백질의 고유한 접착 능력을 통하여 수술 후 부위에 도포하거나 암 부위에 직접 주입하여 국소부위에 광열 치료제를 고농도로 유지시키며 효율적인 항암 치료 효과를 보일 수 있을 것으로 기대된다. 다음으로, 암의 전이를 예방하고 잔류 암세포를 제거하기 위하여 전신적 투여가 가능한 아네로인 기반 나노입자를 개발하였다. 아네로인 기반 나노입자는 전기방사를 진행하면서 빛을 가하여 빠르게 다이타이로신 결합을 형성시켜 제작되었으며, 균일한 입자 크기를 가지는 것을 확인하였다. 세포 독성실험을 통하여 아네로인 나노입자의 생체적합성을 증명하였으며, MCF7 세포를 BALB/c 누드 마우스에 주입한 유방암 종양 모델을 통하여 아네로인 나노입자가 EPR 효과를 통하여 암세포에 잘 전달됨을 확인함으로써 정맥주사를 통한 광열 치료제로서의 활용 가능성을 보았다. 종합적으로 본 학위 논문을 통해 최소한의 부작용으로 항암 치료 능력을 극대화 하기 위하여 두 가지 방식의 해양생명체 유래 단백질 기반 나노입자 시스템을 구성하였다. 홍합 전달 단백질과 아네로인 모두 우수한 생체적합성 및 생분해성을 보유하고 있으며, 본 연구실의 분자 생물 공학 기술을 통하여 균일한 단백질 생산뿐 아니라 변형도 용이하다는 점과 타이로신기를 충분히 보유하고 있어 가교 역시 용이하게 진행 될 수 있다는 장점을 가지고 있어 나노 입자들의 주 재료로 활용되었다. 또한, 안정한 다이타이로신 결합을 통해 나노입자의 안정성이 확보되어 오랜 시간 동안 ICG를 방출하지 않고 보호할 수 있어 광열 치료에 응용될 수 있음을 확인하였다. 본 학위 논문에서 개발된 해양생명체에서 유래한 단백질 기반의 나노입자는 우수한 생체적합성과 생분해성을 가질 뿐 아니라, 홍합접착단백질의 탁월한 수중 접착성과 아네로인의 우수한 물리적 특성을 통해 국소적, 전신적 암 치료에 두루 활용될 수 있을 것으로 크게 기대된다. 홍합접착단백질 기반의 나노입자의 경우는 ICG를 탑재하고 이에 따른 온도 상승효과를 in vitro까지 확인하였기 때문에, 후에 in vivo 상에서도 다음과 같이 종양내 주사나 수술 후 스프레이 기법 두 가지 방법으로 적용될 수 있을 것으로 기대된다. 한편 이번 연구에서 나노입자 제작의 최적화까지만 진행된 아네로인의 경우에는, 광열제인 ICG를 탑재하고, EPR effect 와 더불어 active targeting agents를 도입함으로써 더욱 확실한 항암 효과를 확인할 수 있을 것이다. 또한, ICG뿐 아니라 dityrosine도 자체의 형광을 가지고 있기 때문에 이 두 물질의 특징을 활용하여 치료와 진단을 동시에 할 수 있는 theragnosis로서의 활용성에 대한 연구도 진행될 수 있을 것으로 기대된다
