Investigation of biological activities of some edible mushroom species

YÖK Tez No: 608411Mevcut tez çalışmasında, önemli biyoaktif bileşenleri yapısında bulunduran yenilebilir mantarlardan Rhizopogon luteolus Fr., Rhizopogon roseolus (Corda) Th. Fr. ve Suillus collinitus (Fr.) Kuntze türlerinden elde edilen aseton ve su ekstraktları kullanılmıştır. İlgili ekstraktlar ile muamele edilen insan periferal lenfosit kültürü üzerinde, toplam antioksidan kapasite (TAK) ve toplam oksidan durum (TOD) düzeyleri incelenmiştir. Bahsi geçen aktiviteler analiz edilirken aynı zamanda ilgili ekstraktların lenfositler üzerinde genetik hasar ve sitotoksik etki oluşturma düzeyleri de test edilmiştir. Genel olarak, ekstraktların hücreler üzerinde meydana getirdiği TAK düzeyleri incelendiğinde, S. collinitus'a ait uygulamalar hariç, konsantrasyona bağlı bir artış gözlenirken, TOD verileri ve konsantrasyon arasında negatif bir korelasyon tespit edilmiştir. Genotoksisite testleri (kromozom aberasyonu ve mikronükleus analizleri) ise ekstrakt uygulamalarının test edilen konsantrasyonlarının insan periferal lenfosit kültürü üzerindeki genotoksisiteyi negatif kontrol uygulamasına kıyasla anlamlı derecede (p > 0.05) değiştirmediğini ortaya çıkarmıştır. Elde edilen tüm sonuçlar ele alındığında, R. luteolus ve R. roseolus ekstraktlarının özellikle 50 ve 100 mg/L konsantrasyonlu uygulamalarının insan bağışıklık sisteminde önemli bir görevi olan lenfositlerin TAK değerini genetik veya oksidatif stres meydana getirmeksizin yükselttiği belirlenmiştir.In the present thesis, acetone and water extracts obtained from edible mushrooms, Rhizopogon luteolus Fr., Rhizopogon roseolus (Corda) Th. Fr. and Suillus collinitus (Fr.) Kuntze, containing important bioactive components were used. Total antioxidant capacity (TAC) and total oxidant status (TOS) levels were examined on human peripheral lymphocyte culture treated with the respective extracts. While analyzing mentioned activities, the levels of genetic damage and cytotoxic effect of the respective extracts on lymphocytes were also tested. In general, when the TAC levels of the extracts on the cells were examined, while a concentration-dependent increase was observed except for S. collinitus applications, a negative correlation was found between TOS data and concentration. Genotoxicity tests (chromosome aberration and micronucleus analysis) revealed that the concentrations of the extract applications did not significantly (p > 0.05) change the genotoxicity on human peripheral lymphocyte culture compared to the negative control. Considering all the results obtained, it was determined that the applications of R. luteolus and R. roseolus extracts, especially at concentrations of 50 and 100 mg/L, increased the TAC value of lymphocytes, which play an important role in the human immune system, without causing genetic or oxidative stress
