Nanoparticles (NPs) have been identified as a promising carrier and adjuvant system in mucosal vaccination. Chitosan is widely used as excipient for NPs for this purpose. To make such NPs applicable as dry powder, they can be incorporated into a microparticulate matrix (Nano-in-Microparticles). For an efficient nasal deposition of those microparticles, they should exhibit a particle size above 10 µm. Upon deposition the microparticulate matrix should release the NPs in their original size. The range of suitable sizes for an uptake of the NP in immune competent cells, especially in dendritic cells, is between 100 nm and 500 nm, as known from literature.
