Chitosan Nanopartikel für Mukosale Impfung: Verständnis des Partikelformungsprozesses und Formulierung für die Nasale Applikation

Nanoparticles (NPs) have been identified as a promising carrier and adjuvant system in mucosal vaccination. Chitosan is widely used as excipient for NPs for this purpose. To make such NPs applicable as dry powder, they can be incorporated into a microparticulate matrix (Nano-in-Microparticles). For an efficient nasal deposition of those microparticles, they should exhibit a particle size above 10 µm. Upon deposition the microparticulate matrix should release the NPs in their original size. The range of suitable sizes for an uptake of the NP in immune competent cells, especially in dendritic cells, is between 100 nm and 500 nm, as known from literature. This work focussed on the preparation and characterisation of the NPs. They were prepared from chitosan and carboxymethylcellulose. Ovalbumin was used as model antigen. The particle formation between the two polymers is based on cross-linking via ionic gelation. Effects on particle stability, particle size and protein encapsulation were tested. NPs in the range between 100 - 500 nm could be produced. The model antigen could be loaded in the NPs, depending on the method used rather high loading efficiencies of up to 60 - 70 % (related to the total protein amount) or loading capacities of up to 50 % (related to the total excipient amount) were achieved. Furthermore, a dosage form for the nasal application of the NPs was developed. For that purpose, they were spray dried with mannitol to get embedded in microparticles. This way the NPs were stabilised and in a suitable size range for nasal deposition of approximately 20 µm. Experiments with a nasal cast model showed the suitability of the formulation for nasal application. First in vitro uptake experiments with redispersed NPs in dendritic cells showed a concentration- and size-dependent uptake. Smaller particles up to 500 nm were internalised in cells while larger particles (1000 nm) stuck to the surface of the cells.Forschung zu mukosaler Impfung hat Nanopartikel (NP) als vielversprechenden Antigenträger mit adjuvanten Eigenschaften identifiziert. Häufig wird Chitosan zur Herstellung der NP verwendet. Um NP als trockenes Pulver applizierbar zu machen, werden sie in eine mikropartikuläre Matrix (Nano-in-Mikropartikel) inkorporiert. Die Mikropartikel sollen über die Nase appliziert werden und daher eine Partikelgröße von über 10 µm aufweisen. Für die Aufnahme der NP in immunkompetente, insb. in dendritische, Zellen eignet sich (entsprechend der Literatur) eine Größe von 100 - 500 nm. Der Fokus dieser Arbeit lag auf der Herstellung und Charakterisierung der NP. Diese wurden aus Chitosan und Carboxymethylcellulose hergestellt. Als Modellantigen wurde Ovalbumin verwendet. Die Partikelbildung zwischen beiden Polymeren beruht auf einer Quervernetzung durch ionische Gelierung. Es wurden Einflüsse auf die Partikelstabilität, -größe und Proteinverkapselung untersucht. NP im gewünschten Größenbereich konnten hergestellt werden. Abhängig von der verwendeten Methode konnte eine hohe Verkapselungseffizienz von bis zu 60 - 70 % (bezogen auf Proteingehalt) oder eine hohe Partikelbeladung von bis zu 50 % (bezogen auf Menge Partikelausgangsstoffe) erreicht werden. Im Weiteren wurde eine Darreichungsform für die nasale Gabe der NP entwickelt. Dafür wurden sie mittels Sprühtrocknung mit Mannitol in Mikropartikel eingebettet. Die Partikel wiesen eine geeignete Größe von ca. 20 µm für die nasale Deposition auf, dies konnte auch in Versuchen mit einem Nasenmodell gezeigt werden.
