The measurement and distribution of ribonuclease in human serum and tissues

Bu çalışmada Ribonükleaz enziminin insan serum ve dokularındaki dağılımı iki farklı substrat kullanılarak araştırılmıştır. Sağlıklı 46 bireyde serum ribonükleaz aktivitesi substrat olarak polisitidilik asid kullanılan yöntemle 17.1+8.8 kil/1, maya RNA' sı kullanılan yöntem ile 2.6±1.3 kU/l bulunmuştur. Enzimin dokulardaki dağılımı incelendiğinde en yüksek enzim ak-tivitesi pankreas dokusunda polisitidilik asid substratına karşı 1164± 531 U/l protein olarak elde edilirken; karaciğer ve dalak dokusunda maya RNA' sına karşı sırasıyla 567±281 ve 517±180 U/mg protein olarak bulunmuştur. Böbrek dokusunda ise her iki metod arasında anlamlı fark saptanmamıştır. Bu sonuçlar serumdaki ribonükleaz aktivitesinin büyük ölçüde pankreas dokusundan kaynaklandığını, farklı substratlar kullanma yoluyla diğer dokulardan kaynaklanan enzim düzeylerinin de saptanabileceğini ve patolojik durumlar hakkında bilgi elde edilebileceğini düşündürmektedir.In this study, the authors'purpose was to investigate the distribution of Ribonuclease in human serum and tissues through using two different of substrates. Ribonuclease activity was found 17.1±8.8 kU/l and 2.6±1.3 kU/l in 46 healthy control subjects of which polycytidilic acid and yeast RNA was used, respectively. In tissues, the highest ribonuclease activity was ob-tained in pancreas as 1164+ 531 U/l protein in the method which used polycitidilic acid as a subtrate, while liver and spleen showed greater activity against yeast RNA as 567±281 and 517±180 U/mg protein, respectively. In kidney, there was no difference between the activities in both of the methods. According to these results, it was postulated that the serum ribonu-clease activity mostly originates from the pancreas and it is possible to evaluate the tissues by measuring the activities of serum ribonuclease against different kinds of substrates in pathological conditions
