Lentiviral γ-globin vectors were initially introduced and are constantly beingimproved as a potentially efficient tool for thalassemia gene therapy. The choice of γ-gene as a therapeutic transgene is based on its anti-sickling effect and its ability topair with the α-globin chains, generating functional HbF. However, their use has oftenbeen hampered by low titers, variable expression and gene silencing. To addressthese issues, previous studies in our lab have successfully developed a series ofoncoretroviral vectors, containing the HPFH-2 enhancer combined with the -117activating Greek mutation of the Aγ-gene promoter and the HS-40 enhancer of the α-globin gene locus, that exhibited high level of Aγ-gene expression with absence ofsilencing. ...
