Study of the influence of soy diet on the ultrastructure of the genital tract of the male rat

Soy products are recommended in human diet because of their beneficial effects in menopause and their potential protective effects in cancer and other diseases. Nevertheless, a concern has been raised among scientists, because of their estrogenic effect. Phytoestrogens contained in soy products bind to estrogen receptors and may disrupt the endocrine system. Thus, they could influence the development of the reproductive system during crucial stages of life like gestation, lactation and early development after birth, and/or they could influence spermatogenesis and reproductive function in adults. The main objective of this study was to test the effect of soy diet on the ultrastructure of male reproductive system in adult rats, as well as in young rats that received diet enriched in soy proteins during gestation, lactation and early development. Six groups of male albino Wistar rats were used: Two groups of young male rats 5 weeks old (groups A and B) and 4 groups of adult male rats 6 months old (groups C, D, E and F). The parents of the young animals comprising the study group (group A) received ad libidum food enriched in soy protein before mating and during gestation and lactation, while the offsprings went on receiving the same food up to the age of five weeks. The control group (group B) and their parents received ad libidum the standard food before and during gestation, during lactation and up to the age of five weeks. The animals of groups C and E received ad libidum food enriched in soy protein for 6 and 12 weeks respectively. The animals of groups D and F received ad libidum the standard food for 6 and 12 weeks respectively. All groups were grown under the same conditions and according to the rules of Good laboratory Practice. The study was approved by the local Ethics Committee. The animals were sacrificed and specimens of the reproductive organs were prepared for optical and electron microscopic examination. The preparations were observed and assessed by two different persons, blinded on the source of the specimen. There was no macroscopic difference between the study and the corresponding control groups on the basic anatomical structure of the reproductive organs. Examined under optical microscope, the seminiferous epithelium, the epididymidal epithelium and the epithelium of the vas deferens and the seminal vesicles was found lower in the study groups compared to the corresponding control groups. In the ultrastructure of the seminiferous epithelium of the study groups, spermatocytes appeared with endocytoplasmic and perinuclear oedema, polycystic mitochondria and rupture of the nuclear membrane. The number of spermatozoa in adult animals was lower in rats fed with soy diet, compared to the rats fed with standard food. Soy diet during gestation, lactation and early development had detrimental effects on the ultrastructure of the seminiferous epithelium of young rats and influenced the number of spermatozoa in adult rats. This effect must be taken into consideration in dietary habits during pregnancy, lactation and early stages of life, as well as when fertility is intended in adults.Σκοπός της εργασίας αυτής ήταν να μελετηθεί η επίδραση της διατροφής με σόγια στην υπερμικροσκοπική δομή του γεννητικού συστήματος του άρρενα επίμυα, τόσο κατά την ενήλικο ζωή, όσο και κατά τις κρίσιμες φάσης της ανάπτυξης του γεννητικού συστήματος, δηλαδή κατά την κύηση, τη γαλουχία και την πρώιμη ανάπτυξη. Χρησιμοποιήθηκαν έξι ομάδες αρσενικών επίμυων της φυλής albino Wistar: δυο ομάδες ανήλικων πειραματοζώων ηλικίας 5 εβδομάδων (ομάδες Α και Β) και τέσσερεις ομάδες ενήλικων πειραματοζώων ηλικίας 24 εβδομάδων (ομάδες Γ, Δ, Ε και Ζ). Οι ομάδες Α, Γ και Ε ήταν οι ομάδες μελέτης και οι ομάδες Β, Δ και Ζ ήταν οι αντίστοιχες ομάδες ελέγχου. Η ομάδα μελέτης Α έλαβε τροφή με βάση το σογιάλευρο κατά την ενδομήτρια ζωή, τον θηλασμό και μέχρι τη θυσία των πειραματοζώων. Η ομάδα ελέγχου Β έλαβε τυπική τροφή κατά το ίδιο χρονικό διάστημα με την ομάδα Α. Η ομάδα μελέτης Γ έλαβε τροφή με βάση το σογιάλευρο επί 6 εβδομάδες. Η ομάδα ελέγχου Δ έλαβε τυπική τροφή επί 6 εβδομάδες. Η ομάδα μελέτης Ε έλαβε τροφή με βάση το σογιάλευρο επί 12 εβδομάδες. Η ομάδα ελέγχου Δ έλαβε τυπική τροφή επί 12 εβδομάδες. Ιστοτεμάχια από τα όργανα του γεννητικού συστήματος μελετήθηκαν στο οπτικό και στο ηλεκτρονικό μικροσκόπιο. Οι διαφορές που παρατηρήθηκαν με το οπτικό μικροσκόπιο ήταν οι εξής: To επιθήλιο στις ομάδες μελέτης Α, Γ και Ε, ήταν κατά τόπους χαμηλό, μονόστιβο ή δίστιβο, και δεν εντοπίζονταν στερεοκροσσοί στην κορυφή του. Ο αριθμός των σπερματοζωαρίων στον αυλό των ορχικών σωληναρίων ήταν μικρότερος στην ομάδα Γ σε σύγκριση με την ομάδα Δ, και ήταν πολύ λιγότερα στην ομάδα Ε σε σύγκριση με την ομάδα Ζ. Τα ευρήματα από το ηλεκτρονικό μικροσκόπιο εντοπίσθηκαν κυρίως στα κύτταρα του σπερματικού επιθηλίου των πειραματοζώων της ομάδας Α που είχαν εκτραφεί με σογιάλευρο κατά την κύηση, τη γαλουχία και κατά την πρώιμη μετεμβρυϊκή ζωή. Στα περισσότερα κύτταρα παρατηρήθηκαν ρήξη και εγκολπώσεις της πυρηνικής μεμβράνης, κατάτμηση της χρωματίνης στον πυρήνα, έξοδος πυρηνικού υλικού και διάχυτο ενδοκυττάριο οίδημα. Στα σπερματοκύτταρα των ομάδων Γ και Ε των ενήλικων πειραματοζώων που είχαν λάβει τροφή με βάση το σογιάλευρο μετά την ενηλικίωση, χαρακτηριστικά ευρήματα ήταν το διάχυτο ενδοκυττάριο οίδημα, η διεύρυνση του λείου ενδοπλασματικού δικτύου, τα οιδηματώδη, κατά τόπους άδεια ακρολοφιών και ενίοτε πολυκυστικά μιτοχόνδρια, και βέβαια ο αριθμός των σπερματοζωαρίων, ο οποίος ήταν μικρότερος σε σύγκριση πάντα με τα πειραματόζωα των αντίστοιχων ομάδων ελάγχου Δ και Ζ. Ιδιαίτερα στα πειραματόζωα της ομάδας Ε που είχαν λάβει σόγια για διάστημα 12 εβδομάδων, ο αριθμός των σπερματοζωαρίων ήταν πολύ μικρός. Σύμφωνα με τα αποτελέσματα της παρούσας μελέτης, φαίνεται ότι η διατροφή με σόγια επηρεάζει την μικροσκοπική και την υπερμικροσκοπική δομή του γεννητικού συστήματος και τον αριθμό των γεννητικών κυττάρων τόσο στους ενήλικους όσο και στους ανήλικους αρσενικούς επίμυες. Επομένως, θα πρέπει να αντιμετωπίζεται με σκεπτικισμό η κατανάλωση υψηλής ποσότητας σόγιας όταν είναι επιθυμητή η εγκυμοσύνη, καθώς και κατά τις κρίσιμες περιόδους της ανάπτυξης, όπως κατά την κύηση, κατά τον θηλασμό και κατά την πρώτη περίοδο μετά τη γέννηση
