Detection of shiga toxin producing Escherichia coli isolated from mastitis milk of sheep by qPCR technique

This study was conducted to determine the Shiga toxin producing Escherichia coli isolated from mastitis milk samples of sheep that collected from Diwaniya city by qPCR technique. A total of (60) clinical investigated mastitis milk samples from sheep were subjected to bacterial DNA extraction by using (PrestoTM Mini gDNA Bacteria Kit). The extracted DNA subjected to Real-Time PCR technique (qPCR) for the detection of Shiga toxin 1 (stx1) gene. Results were revealed that the mastitis sheep are more prevalence for shedding Shiga toxin 1 producing Escherichia coli (13.3%) of (8/60) positive samples. In conclusion, the Real-Time PCR technique was shown high specific and rapid method in detection of Shiga toxin gene and the sheep which infected by mastitis can be carried the shiga toxin producing Escherichia coli, that may be considered to be risk factor of human infection
