We tracked in vitro replication of each Env281/375 polymorphism for (A) CAP224, (B) CAP304, and (C) CAP174 by measuring p27 levels in activated primary rhesus CD4+ T cells over the course of 14 days. Individual SHIVs were used to infect 106 activated rhesus CD4+ T cells at a MOI of 0.01. Culture supernatants were collected over 14 days, with media replacement at each collection time-point to maintain cultures in 2 mL total volume. Viral p27 protein was quantified by ELISA. Env375 mutants contain a histidine except for CAP224 which has a tryptophan mutation. Wildtype 281 virus cultures are denoted (*).</p
