Titin consists of different functional units that provide elasticity and support sarcomere assembly. The presence of a kinase domain and binding sites for structural and signaling proteins suggest an important role during myofibrillogenesis and as biochemical stress sensor. To elucidate the function of Titin s M-line in early cardiac development a constitutive Titin MEx1/2 knockout was generated. KO embryos develop normally until E9.0 apparent through a contracting heart and a regular number of somites. From E9.5 development is delayed indicated by a reduced body size, decreased trabeculation of the heart ventricle, and a thin myocardium. They fail to thrive and cardiac dysfunction causes lethality by E11.5. Ultrastructural analysis confirms initial sarcomere assembly at E9.0 but impaired lateral growth. From E10.5 sarcomeres disassemble and only few filaments in disarray remain at E11.0. Immunocytochemistry shows integration of Titin into the Z-disc and at the A/I-junction. Titin s M-line region does not integrate into the M-band of KO cardiac muscle cells suggesting the formation of a continuous Myosin but a discontinuous Titin filament. Nevertheless the structural protein Myomesin-EH integrates into the M-band although its binding site to Titin is deleted in the KO. The failure of Myomesin to cross-link Titin in the M-band results in increased mobility of the sarcomeric filaments. Thus enhanced mechanical strain on the growing sarcomere leads to its disassembly. Titin s kinase domain has been implied in sarcomere assembly and the control of muscle gene expression through the in vitro substrates Nbr1, Sqstm1, and T-cap. This study shows that the phosphorylation of T-cap by Titin s kinase is not the regulatory principle for initial cardiac sarcomere assembly. Not only do sarcomeres form in the absence of Titin s kinase, but furthermore T-cap is not detectable in early sarcomere development. Moreover, the substrates Nbr1 and Sqstm1 were not expressed in significant amounts at the time when the knockout phenotype develops. Titin s M-line region also contains binding sites for ubiquitin ligases. MuRF-1 is involved in protein degradation in the atrophic skeletal muscle and MuRF-2 regulates gene expression by acting as biochemical stress sensor. In this study both proteins are highly expressed at the time when the knockout phenotype develops. But neither the absence of Titin s M-line nor the atrophy altered the expression of MuRF-1 and MuRF-2. However, differences in the expression of sarcomeric proteins in the embryonic versus the adult heart refer to different molecular mechanisms of sarcomere assembly and maintenance of preexisting structures. This study provides in vivo data indicating that Titin s M-line is dispensable for initial sarcomere assembly but required for lateral growth and stabilization of the sarcomere. Furthermore it was shown that structural, mechanical, and signaling functions of Titin and its interacting proteins in the embryonic sarcomere are different from those observed in adult. The mouse model helps to understand how Titin and its binding protein shape and regulate sarcomere assembly in early cardiac development. The insights into the pathophysiology and molecular mechanism of cardiomyopathies can be used to develop new therapeutic strategies for cardiovascular diseases.Titin s M-Linie ist wichtig für die Reifung der Myofibrillen und Kontraktilität des Herzens Die kleinste kontraktile Einheit der quergestreiften Muskulatur ist das Sarkomer. Es besteht aus Myosin, Aktin und dem elastischen Titin, die ein kontinuierliches Filamentsystem formen. Titin ist aus unterschiedlichen funktionellen Untereinheiten aufgebaut und sorgt für Elastizität, Aufbau und Stabilisierung des Sarkomers. Um die Bedeutung von Titins Kinase Domäne als biomechanischen Stresssensor und von Titins M-Linie während der Myofibrillogenese aufzuklären, wurde ein Titin Mex1/2 Knockout- Mausmodell hergestellt. Knockout Embryonen entwickelten sich bis E9.0 morphologisch unauffällig. Ab E9.5 war jedoch die Entwicklung gestört. Die KO Embryonen waren etwas kleiner, zeigten eine geringere Trabekulierung des Herzventrikels und ein dünneres Myokard. Die Pathophysiologie führte zum Herzstillstand und Tod der KO Embryonen bis zum Tag E11.5. Ultrastrukturanalysen zeigten eine initiale Assemblierung der Myofibrillen bis E9.5 aber anschließend ein gestörtes Dickenwachstum und Disassemblierung der Myofibrillen bis E11.5. Anhand von Immunofärbungen konnte gezeigt werden, dass das defekte Titin in die Z-Scheibe, I-Bande und teilweise A-Bande aber nicht in die M-Bande integriert. Daher kommt es zur Ausbildung eines kontinuierlichen Myosinfilamentes aber nicht zu einem kontinuierlichen Titinfilament. Das M-Banden Strukturprotein Myomesin-EH integrierte in die M-Bande obwohl die Bindungsstselle im defekten Titin fehlt. Titins Kinase hat potentielle in vitro Substrate wie Nbr1, Sqstm1 und T-cap, die sowohl an der Regulation von Muskelproteinexpression als auch am Aufbau der Sarkomere beteiligt sein sollen. Zum ersten mal konnte gezigt werden, dass die Phosphorylierung von T-cap durch die Kinase kein entscheidender Mechanismus während der Myofibrillogenese ist, da sich auch in Abwesenheit der Kinase Sarkomere bilden und T-cap erst während der späten embryonalen Entwicklung expremiert wird. Auch Nbr1 und Sqstm1 sind zu der Zeit der Enstehung des Knockout-Phänotypes noch nicht expremiert. Die E3 ubiquitin Ligasen MuRF-1 und 2 interagieren auch mit Titins M-Linie. MuRF-1 erhöht die Proteindegradation bei Atrophie und MuRF-2 stabilisert die M-Bande und reguliert die Expression von Muskelproteinen. Beide Proteine waren zwar während der frühen embryonalen Entwicklung stark expremiert aber ein Unterschied in der Expression von Knockout und Wildtyp konnte nicht festgestellt werden. Daher kann man sagen, dass der Unterschied in der Expression von Muskelproteine im embryonalen und adulten Herzen aufgrund der unterschiedlichen molekularen Mechanismen vom Aufbau des Sarkomers und der Erhaltung der bereits bestehenden Sarkomere entsteht. Diese Arbeit liefert in vivo Daten, dass der Aufbau und Kontraktilität des Sarkomers unabhängig von einem kontinuierlichen Titinfilament sind jedoch Titins M-Linie das Sarkomer unter mechanischer Belastung stabilisiert. Diese Mausmodell trägt zum Verständnis der Entstehung der Myofibrillen im Herzmuskel bei und gibt Hinweise zur Pathophysiologie von Kardiomyopathien
