Endogenous GAC63 is important for efficient transcriptional activation by LEF1

After 72 h, cells were harvested for either immunoblots or total RNA preparation. mRNA was analyzed by RT-QPCR to measure the levels of human GAC63, cyclin D1 and β-actin transcripts. Results shown for GAC63 and cyclin D1 transcripts were normalized by the levels of β-actin transcripts and are representative of three independent experiments. A paired, two-tailed -test performed on the values from the three experiments indicated that the siRNA against GAC63 caused significant decreases in the expression levels of cyclin D1 mRNA ( = 0.0004 for 1 and = 0.0008 for #2). For the three experiments, the mean decrease and 95% confidence intervals were 85 ± 5% (#1) and 85 ± 7% (#2) for GAC63 mRNA, and 46 ± 12% (#1) and 51 ± 7% (#2) for cyclin D1 mRNA.
