Western blot analysis of the insulin-signaling cascade in homogenized whole-brains from male animals.

<p> Panel A shows representative Western blots from four DMSO-control and four wortmannin-treated male <i>Pcmt1^−/−</i> animals. The polypeptide molecular weight of each of the bands as determined in comparison with rainbow markers was consistent with the known weight of the target protein (<a href="http://www.plosone.org/article/info:doi/10.1371/journal.pone.0046719#pone-0046719-t001" target="_blank">Table 1</a>). Each row represents an independent exposure. Panel B gives the results of densitometry from the entire group of 7 control and 11 wortmannin-treated animals. All bands were standardized to GAPDH controls to ensure equal protein loading. The error bars denote standard deviations; asterisks indicate a statistical significance (p<0.05) by Student t-test between the wortmannin and DMSO control samples.
