RNA and protein expression of human P-gp in control cells (Flp-In™-293 cells), cells expressed wild-type P-gp and cells expressed variant-type P-gp.

<p>(<i>a</i>) RNA expression was detected by real-time quantitative RT-PCR. There was no significant different RNA expression among cells expressed wild-type P-gp and cells expressed variant-type P-gp, whereas little <i>ABCB1</i> gene expression was detected in the control cells. Data were presented as mean ± SE of at least three experiments, each in triplicate.(<i>b</i>) Western blot of P-gp expression with C219 monoclonal antibody (2 µg protein/lane). Arrows marked the mature fully glycosylated (∼170 kD) and immature P-gp (∼150 kD). The expression of β-actin was used as the loading control. No significant difference was detected among cells expressed wild-type P-gp and cells expressed variant-type P-gp.
