Apskatīti metodes validācijas galvenie parametri. Darba eksperimentālajā daļā veikta spektrofotometriska orotskābes noteikšana, izmantojot standartpiedevu metodi. Noteikts orotskābes saturs laboratorijas kontroles paraugā, optimizējot vides pH ietekmi uz analīzes rezultātiem. Eksperimentāli izvērtētas AEŠH metodes purīnu un pirimidīnu noteikšanai cilvēka urīnā. The paper compiles literature review of purines and pyrimidines, they role in human organism, purines and pyrimidines metabolic pathways, enzymes and disorders, of inborn errors of metabolism. Several purines and pyrimidines detection methods were reviewed and the most important parameters of method validation were considered. Experimental part of thesis include orotic acid spectrophotometric determination. Quantitative analysis of orotic acid has been performed using standard addition method. The mean concentration of orotic acid was calculated in pooled control sample and optimal pH value of reaction was found. The HPLC method for purines and pyrimidines quantification in human urine samples has been developed. The HPLC method validation was made according method validation guidelines
