Isolation of Bacteriophages and Their Application to Control Pathogenic Bacteria: Bakteriofag untuk Mengontrol Bakteri Patogen

Foodborne disease (FBD) merupakan suatu fenomena keracunan makanan yang disebabkan oleh bakteri patogen yang terkandung di dalam makanan. Penyakit yang disebabkan oleh FBD dapat berupa penyakit ringan seperti diare hingga penyakit berbahaya yang dapat berujung pada kematian. Salah satu upaya dalam menanggulangi adanya fenomena FBD adalah dengan menggunakan antibiotik sebagai agen kontrol bakteri patogen. Dampak negatif akibat penggunaan antibiotik adalah adanya kemampuan bakteri patogen untuk resisten terhadap antibiotik. Penggunaan bakteriofag yang spesifik terhadap bakteri patogen menjadi salah satu alternatif untuk penggunaan antibiotik. Penelitian ini bertujuan untuk mendapatkan bakteriofag yang dapat menghambat bakteri patogen. Isolasi bakteriofag dilakukan dari beberapa sampel yakni air sampah, usus ayam segar, sayur asin hingga tanah. Hasil penelitian menunjukkan bahwa isolasi bakteriofag menghasilkan bakteriofag campuran (phage cocktail) bersifat virulen dari sampel usus ayam dengan isolat nomor U14 (ՓU14). ՓU14 mampu melisiskan bakteri inang pada penambahan menit ke-30, 60, 90, dan 120. ՓU14 dengan konsentrasi 1,05 x 108 PFU/mL efektif menghambat pertumbuhan bakteri patogen Salmonella typhimurium, Listeria monocytogenes, Staphylococcus aureus dan Bacillus cereus tetapi tidak dapat menghambat pertumbuhan Eschericia coli.Foodborne disease (FBD) is a phenomenon of food poisoning caused by pathogenic bacteria. Diseases caused by FBD can be in the form of mild diseases such as diarrhea or dangerous diseases that can lead to death. An alternative effort to overcome the phenomenon of FBD is by using antibiotics as a pathogenic bacterial control agent. The negative impact of using antibiotics can evoke pathogenic bacteria's ability to resist antibiotics. The use of bacteriophages specific to pathogenic bacteria has become one of the alternatives to eliminate the use of antibiotics. This research aims to obtain bacteriophages that can inhibit pathogenic bacteria. Bacteriophage isolation was carried out from several samples: wastewater, fresh chicken intestines, salted vegetables, and soil. The results showed that the isolation of bacteriophages produced virulent mixed bacteriophages from chicken intestinal samples with isolate number U14 (Փ U14). Փ U14 can lyse all of the host bacteria in the addition of 30 minutes, 60, 90, and 120. Փ U14 with a concentration of 1.05 x 108 PFU / mL effectively inhibits the growth of pathogenic bacteria such as Salmonella typhimurium, Listeria monocytogenes, Staphylococcus aureus, and Bacilluscereus but cannot inhibit growth Escherichia coli.
