ACHILLEA MILLEFOLIUM L. EKSTRELERININ TOPLAM FENOLIK VE FLAVONOIT MADDE KOMPOZISYONLARININ VE ANTIOKSIDAN ETKILERININ BELIRLENMESI

Dünya genelinde ve Türkiye’de çeşitli tıbbi amaçlarla kullanımı bilinen bitkilerden biri olancivanperçemi (Achillea millefolium L.) Compositae familyasına dahildir. Achillea millefoliumantioksidan, antimalaryal, antiparazitik, antimikrobiyal, antifungal, antihipertansif,vazodilatör ve bronkodilatör aktiviteleri olan bir türdür. Bu çalışmada bu bitkinin on farklıekstresi hazırlanarak (%10-%99.9 etanol) bu ekstrelerin toplam fenolik, toplam flavonoitiçerikleri ve ayrıca antioksidan aktiviteleri değerlendirilmiştir. Toplam fenolik ve toplamflavonoit içeriği en yüksek %90 etanol ekstresi bulunmuştur (146,24±0,80 mgGAE/gekstre,62,73±1,26 mgCA/gekstre).Antioksidan aktivite DPPH ve ABTS radikallerini süpürücü özellikleri üzerindendeğerlendirilmiştir. DPPH radikalini süpürücü aktivitede ekstreler 0,00095-1 mg/mLaralığında 11 farklı konsantrasyonda çalışılmıştır. %60-%99.9 aralığında etanol yüzdesinesahip ekstrelerin daha polar olan ekstrelere göre radikali daha güçlü inhibe ettikleribelirlenmiştir. ABTS radikali süpürücü aktivitede ekstreler 0,25 ve 0,5 mg/mLkonsantrasyonda çalışılmış olup %70 etanol ekstresi 0,5 mg/mL konsantrasyonda 2,29±0,16mmol/L Trolox değeriyle en güçlü aktivite sergileyen ekstre olarak değerlendirilmiştir.Geleneksel tıpta uzun yıllardır kullanılan bu türün farklı polaritedeki ekstrelerinin kimyasaliçerikleri ve antioksidan aktiviteleri değerlendirilmiş olup çalışmalarımız farklı biyolojikaktivitelerinin incelenmesi yönünde devam etmektedir.Yarrow (Achillea millefolium L.), which is one of the plants known to be used for variousmedicinal purposes in the world and Turkiye, belongs to the Compositae family. Achilleamillefolium is a species that has antioxidant, antimalarial, antiparasitic, antimicrobial,antifungal, antihypertensive, vasodilator, and bronchodilator activities. In this study, tendifferent extracts of this plant (10% - 99.9% ethanol) were prepared and the total phenolic andflavonoid contents and their antioxidant activities were evaluated. The highest total phenolicand total flavonoid content was found in 90% ethanol extract (146.24±0.80 mgGAE/gextract,62.73±1.26 mgCA/gextract).Antioxidant activity was evaluated on DPPH and ABTS radical scavenging properties. InDPPH radical scavenging activity, the extracts were studied at 11 different concentrations inthe range of 0.00095-1 mg/mL. It was determined that the extracts with an ethanol percentagein the range of 60%-99.9% inhibited the radical more strongly than the more polar extracts. InABTS radical scavenging activity, the extracts were studied at 0.25 and 0.5 mg/mLconcentrations, and the 70% ethanol extract was evaluated as the extract exhibiting thestrongest activity with a Trolox value of 2.29±0.16 mmol/L Trolox at a concentration of 0.5mg/mL. The chemical contents and antioxidant activities of different polarity extracts of thisspecies, which have been used in traditional medicine for many years, have been evaluatedand our studies continue to examine different biological activities.
