An integrative approach to study the seasonality and biodiversity of benthic diatoms in the Adriatic and Tyrrhenian Seas

Il microphytobenthos (MPB) è una componente poco esplorata dell'ecosistema marino nonostante il suo elevato contributo alla produttività primaria e il ruolo chiave nella catena trofica e nei cicli biogeochimici. Questo studio mira a incrementare le conoscenze sulla biodiversità e sulla struttura di comunità delle diatomee bentoniche del Mar Mediterraneo in un ambiente subtidale. Nella prima parte della tesi, è stato studiato l'andamento stagionale delle abbondanze e della composizione specifica delle diatomee bentoniche in due stazioni dell’Adriatico settentrionale mediante microscopia ottica. Sono stati osservati 118 taxa. In entrambe le stazioni le diatomee bentoniche hanno mostrato una spiccata stagionalità, con massima abbondanza, biomassa e biodiversità in primavera e minima in estate. In entrambe le stazioni le forme mobili erano la componente più importante della comunità a diatomee seguite dalle adnate. Il plocon era meno abbondante delle altre forme di crescita, ma in autunno contribuiva in modo significativo alla biomassa. Nella seconda parte di questo studio, 59 ceppi di diatomee sono stati isolati dai sedimenti raccolti presso 2 stazioni nell'Adriatico settentrionale e 11 stazioni nel Mar Tirreno per la caratterizzazione morfologica e molecolare. Tutti i ceppi sono stati osservati e fotografati al microscopio ottico e le loro sequenze del gene 18S RNA sono state determinate e collocate in un albero filogenetico contenente 541 sequenze parziali del gene 18S rRNA. Quindi 13 ceppi scelti tra quelli con una posizione dubbia sono stati analizzati al SEM e TEM per una dettagliata analisi ultrastrutturale e per l’identificazione delle specie. Questo studio ha evidenziato la mancanza di dati relativi alle sequenze per molti taxa e la necessità di utilizzare un approccio integrato, basato sia sull'analisi morfologica sia su quella molecolare, per migliorare le conoscenze tassonomiche e il sistema di classificazione delle diatomee bentoniche e per una migliore comprensione della loro ecologia.Microphytobenthos (MPB) is a poorly explored component of marine ecosystem despite their high contribution to primary productivity and the main role in the trophic chain and in biogeochemical cycles. This study aims to increase the knowledge on the biodiversity and community structure of the subtidal benthic diatoms of the Mediterranean Sea. In the first part of the thesis, seasonal abundance and composition of benthic diatoms was studied at two stations of N Adriatic coast using light microscopy. A total of 118 taxa were observed. At both stations, benthic diatoms showed a marked seasonality, with maximum abundance, biomass and biodiversity in spring and minimum in summer. At both stations the motile forms were the most important component of the diatom communities followed by the adnate ones. The plocon forms were less abundant than the others but in autumn they contributed significantly to the biomass. In the second part of this study, 59 diatom strains were isolated from sediments collected at two stations located in the northern Adriatic and 11 stations in the Tyrrhenian Sea for morphological and molecular characterization. All strains were observed and imaged in light microscopy. Their 18S RNA gene sequences were determined and placed in a phylogenetic tree containing 541 partial sequences of the 18S rRNA gene. Then 13 strains selected among those having a questionable position were analyzed at SEM and TEM for detailed ultrastructural analysis and species identification. This study highlighted the lack of data for many taxa and the need to use an integrative approach, based on both morphological and molecular analysis, to improve the taxonomy and classification system of benthic diatoms and for a better understanding of their ecology
