Liver tumors with activating mutations in the Ctnnb1 gene, encoding beta-catenin, were chemically induced in mice by application of a single dose of the liver carcinogen N-nitrosodiethylamine followed by chronic treatment with the tumor promoter phenobarbital. To further assess the role of beta-catenin in hepatocarcinogenesis, ablation of beta-catenin was accomplished in established Ctnnb1-mutated liver tumors. The results showed that beta-catenin is essential for the regulation of murine liver tumor cell proliferation. The reduction of membranous Connexin 32, a gap-junction forming protein, could be a mechanism to trigger cell proliferation by isolating tumor cells from growth-restraining signals coming from normal neighboring cells.
