Physiological and Molecular Characterization of the Effects of NEP1-like Proteins (NLPs) on Plants

NEP1-ähnliche Proteine oder NLPs (NEP1-like Proteins) wurden nach dem ersten identifizierten Protein dieser Art benannt, NEP1 (Necrosis- and ethylene-inducing peptide 1) aus Fusarium oxysporum. Erstmals beschrieben wurden diese Proteine aufgrund ihrer Eigenschaft, in zweikeimblättrigen Pflanzen Reaktionen der pflanzlichen Immunabwehr auszulösen. Die NLP-Familie umfasst Mitglieder aus Oomyzeten, Pilzen und Bakterien. Nekrotisierende Formen wurden bislang nur in Pathogenen mit nektrotropher oder hemibiotropher Lebensweise identifiziert. Diese Arbeit zeigt, dass NLPs als generelle Virulenzfaktoren von Pilzen, Bakterien und Oomyzeten bezeichnet werden können, da neben virulenzsteigernden Eigenschaften von NLPs aus Pilzen und Bakterien solche auch für NLPs aus Oomyzeten nachgewiesen werden konnten. Dies gelang durch Komplementation eines NLP-defizienten Pectobacterium carotovorum-Stammes mit NLPs aus Oomyzeten. Die virulenzfördernde Eigenschaft von NLPs ist von der nekrotisierenden Aktivität der Proteine und damit von ihrer zytolytischen Aktivität abhängig, wie die Korrelation dieser Aktivitäten nach Analyse von NLP-Punktmutanten zeigt. NLP-induzierte Nekrosen sind daher sehr wahrscheinlich keine Abwehrreaktion, sondern ein durch NLPs verursachtes Krankheitssymptom. NLPs können deshalb als virulenzsteigernde, zytolytische Toxine bezeichnet werden, die den hemibiotrophen bzw. nekrotrophen Lebensstil unterstützen. Die Induktion abwehrspezifischer Markergene ausschließlich durch zelltodvermittelnde NLP-Mutanten zeigt außerdem, dass die Toxinwirkung vermutlich die Grundlage für die Aktivierung pflanzlicher Immunreaktionen ist. Wie in Tieren scheint also auch in Pflanzen der Mechanismus der toxinvermittelten Immunität zu existieren, vermutlich durch die toxinbedingte Generierung von DAMPs. Untersuchungen zum molekularen Mechanismus ergaben, dass NLPs die Integrität der Plasmamembran von Dikotyledonen nach Interaktion mit der äußeren Seite der Membran beeinflussen. Aufgrund der strukturellen Homologie zu Aktinoporinen wird vermutet, dass NLPs hier nach Kontakt mit spezifischen Strukturen Poren bilden und damit das Membranpotential oder die Stabilität der Plasmamembran stören. Biochemische Studien lieferten Hinweise, dass diese NLP-Zielmoleküle Lipidstrukturen sind. Die funktionelle Charakterisierung hochkonservierter Aminosäuren im Vergleich mit der Kristallstruktur von NLPPya ergab, dass für die Toxinaktivität der NLPs in einer putativen Bindestelle befindliche Aminosäuren essentiell sind. Diese Aminosäuren sind an der Koordinierung eines divalenten Kations, vermutlich Kalzium, beteiligt. Dieses scheint demnach wichtig für die Aktivität von NLPs zu sein und könnte der spezifischen Bindung an NLP-Zielmoleküle dienen.NEP1-like proteins (NLPs) were named after the first identified protein of this family, NEP1 (Necrosis- and ethylene-inducing peptide 1) from Fusarium oxysporum. These proteins were first described due to their ability to trigger cell death and immune responses in dicotyledonous plants. The NLP family comprises members from oomycetes, fungi and bacteria. So far, necrotizing NLPs were only identified in pathogens with a necrotrophic or hemibiotrophic life style. This work shows that NLPs can be designated as general virulence factors since virulence promoting characteristics were demonstrated not only for fungal and bacterial NLPs but also for oomycete NLPs. This was achieved by complementation of a NLP-deficient Pectobacterium carotovorum strain with oomycete NLPs. The virulence promoting property of NLPs is dependent on its necrotizing and cytolytic activity, shown by a correlation of these attributes after an analysis of mutated NLP versions. Hence, NLP-triggered necrosis is probably not a defense reaction, but a disease symptom. NLPs can be considered virulence-promoting cytolytic toxins supporting the hemibiotrophic or necrotrophic lifestyle of pathogens. The induction of defense-related marker genes exclusively by cell death-inducing NLP mutants shows, that the toxin activity itself seems to cause the onset of plant immune responses. Thus, like in animals, a mechanism of toxin-induced immunity in plants can be assumed, probably by a toxin-mediated generation of DAMPs. Research on the molecular mechanism of NLP action showed, that following a contact with the outer site of the plasma membrane of dicots, NLPs affect the membrane integrity. Due to structural homologies with actinoporins, a pore forming mechanism after contact with membrane-specific structures is hypothesized with a following disturbance of the membrane potential or the membrane stability. Biochemical studies pointed to lipid-like attributes of the target molecules. The functional analysis of highly conserved amino acids compared to the crystal structure of NLPPya emphasized the crucial functions of amino acids that are located in a putative binding pocket and are involved in the coordination of a bivalent cation, presumably calcium.
