Comparative studies to Salmonella detection from food with ISO 6579:2002 having regard to chromogenic culture media

Infektionen durch Enteritis-Salmonellen gehören zu den am häufigsten erfassten Ursachen von Durchfallerkrankungen bei Menschen in den westlichen Industrienationen und werden überwiegend durch den Verzehr von kontaminierten Lebensmitteln tierischen Ursprungs ausgelöst. Weltweit ist durch enorme Fortschritte auf den Gebieten der Tierseuchen- und Krankheitsbekämpfung sowie der Lebensmittelhygiene ein steter Rückgang der Salmonellose des Menschen zu erkennen, dennoch wird sie auch zukünftig eine Gefährdung der menschlichen Gesundheit darstellen. Die kulturelle Referenzmethode zum Nachweis von Salmonellen in Lebensmitteln umfasst eine nicht-selektive Voranreicherung, gefolgt von einer Selektivanreicherung mit anschließender Subkultivierung auf feste Selektivnährböden mit einer vorläufigen biochemischen und serologischen Bestätigung. Das Arbeitsprotokoll dieses sehr zeit- und materialaufwendigen Nachweisverfahrens wird in regelmäßigen Abständen von den Normierungskomitees überprüft und überarbeitet. In der vorliegenden Arbeit sollte der aktuelle und umstrittene Änderungsentwurf der horizontalen Norm zum Nachweis von Salmonellen aus Lebensmitteln und Futtermitteln ISO/DIS 6579:2000 mit dem noch geltenden Referenzverfahren nach § 64 LFGB kritisch verglichen werden. Gleichzeitig sollten die neuen chromogenen Selektivnährmedien AES-Salmonellen-Agar-Platte (ASAP), Oxoid Salmonella Chromogen-Medium (OSCM) und Miller-Mallinson-Agar (MM) hinsichtlich ihrer Effizienz geprüft werden. Von 286 nativen Hackfleisch- und Geflügelfleischproben waren insgesamt 39 (13,6 %) mit Salmonellen kontaminiert. Die höchste Nachweisrate gelang mit einer Selektivanreicherung in RVS (97,4 %), gefolgt von MKTTn (94,9 %) und SC (38,5 %). Im Vergleich zu den herkömmlichen Nährmedien BPLS, XLD und XLT4 gelang mit den neuen chromogenen Nährmedien MM, ASAP und OSCM bereits nach verkürzter Anreicherungszeit in RVS eine höhere Nachweisrate. Statistisch betrachtet konnte zwischen der alten und der neuen Referenzmethode kein signifikanter Unterschied gezeigt werden. Demnach ist die neue Referenzmethode als gleichwertig anzusehen. Sowohl unabhängig von der eingemischten Salmonella-Konzentration von 1-10, 11-50 oder 51-200 KbE S. Typhimurium/25 g Hackfleisch als auch von den im Versuch eingesetzten festen Selektivnährmedien, konnte bereits nach 24-stündiger Selektivanreicherung in RVS mit 24-stündiger Subkultivierung die höchste Salmonella-Wiederfindungsrate von jeweils 100 % erzielt werden. Nur mit dem MM-Medium gelang der Nachweis charakteristischer Salmonella-Kolonien sowohl von den ausgewählten Prüfstämmen der Serovare S. Senftenberg, Salmonella arizonae, S. Dublin als auch von S. Derby. Drei Non-Salmonella-Prüfstämme führten zu falsch-positiven Ergebnissen auf BPLS, XLD und XLT4. In eigenen Untersuchungen konnte ein repräsentatives Serovar- und Resistenzspektrum aus nativen Lebensmittelmatrizes im Laufe eines Jahres isoliert werden. Aus 39 Salmonella-positiven Hackfleisch- und Geflügelfleischproben konnten 43 Salmonellen-Stämme isoliert werden. Es dominierte der Serovar S. Typhimurium (n = 21) mit den Subtypen DT 104 B low, DT 104 L, DT 012 und RDNC. Einzelne Stämme der Serovare S. Paratyphi B d-Tartrat-positiv und S. Saintpaul, die aus der Matrix ´Geflügelfleisch isoliert wurden, waren multiresistent.Infections with nontyphoid Salmonellae are one of the most commonly recorded cause of gastroenteritis in humans in the Western industrial nations. The great majority of human cases of salmonellosis are due to the consumption of contaminated foods of animal origin. An enormous progress in the areas of epidemic and animal disease control as well as food hygiene has been a visible reduction of food borne salmonellosis worldwide but it will still be a risk to human health in the future. The golden standard for the detection of Salmonella from food relies on a nonselective preenrichment, followed by a selective enrichment step, isolation on selective agar media and a preliminary biochemical and serological confirmation. This conventional cultural method is very time consuming and expensive thus its protocol is always revised by the standard committees. In this study the disputed draft ISO/DIS 6579:2000 of the horizontal method for the detection of Salmonella from human food and animal feed was compared critically with the standard method according to § 64 LFGB. The efficiency of three new chromogenic plating media, AES Salmonella Agar plate (ASAP), Oxoid Salmonella Chromogen Medium (OSCM) and Miller Mallinson Medium (MM) was also tested. About 39 (13,6 %) of 286 naturally contaminated samples of minced meat and fresh poultry were Salmonella spp. positive. RVS broth yielded the highest recovery of Salmonella spp. and was superior (97,4 %) to MKTTn broth (94,9 %) and SC broth (38,5 %). After inoculating for 24 hours only in RVS broth the new chromogenic plating media MM, ASAP and OSCM produced the best results compared with BPLS, XLD and XLT4. No significant difference was obtained by comparing the ISO/DIS 6579:2000 draft to the conventional cultural method. Therefore, the draft is no less effective than the conventional method. A 24-hours-enrichment in RVS broth yielded a Salmonella recovery of 100 % from artificially contaminated minced meat (1-10 cfu/11-50 cfu/51-200 cfu S. Typhimurium into 25 g). Only MM could detect atypical Salmonella test strains of S. Senftenberg, Salmonella arizonae, S. Dublin and S. Derby. BPLS, XLD and XLT4 failure to detect three non-Salmonella test strains. In own investigations a representative spectrum of common serotypes and antimicrobial drug resistance was isolated from food samples during the course of one year. From 39 naturally contaminated minced meat and fresh poultry samples 43 Salmonella strains could isolated. S. Typhimurium (n = 21) with the subtypes DT 104B low, DT 104 L, DT 012 and RDNC prevailed.
