ANTI GENOTOKSIK RESVERATROL TERHADAP BONE MARROW MENCIT (Mus musculus) SWISS WEBSTER YANG DIINDUKSI ALUMINIUM KLORIDA (AlCl3): ANALISIS MN ASSAY

Akumulasi aluminium dalam bentuk ion Al3+ pada tubuh yang banyak ditemukan dalam kehidupan sehari-hari dengan jangka waktu selama 7 tahun dapat menyebabkan toksik. Hal ini disebabkan ion Al3+ mempunyai afinitas yang kuat dibandingkan dengan besi (Fe) untuk berikatan dengan transferrin sehingga menghambat pengikatan oksigen di dalam darah dan menyebabkan produksi Reactive Oxygen Species (ROS). Hal ini menimbulkan patahan DNA yang diindikasikan oleh adanya mikronukleus. Dalam mengurangi terjadinya produksi ROS berlebih, dibutuhkan bahan alam yang terdapat gugus stilbenoid yang berikatan dengan senyawa oksidan dan radikal bebas yaitu resveratrol. Di Indonesia, resveratrol telah berhasil diisolasi dari tempe, tetapi perlu adanya penelitian in vivo tentang efek anti genotoksiknya. Metode penelitian yang digunakan adalah Rancangan Acak Lengkap (RAL) dengan 4 perlakuan, yaitu kelompok kontrol, kelompok AlCl3 dengan dosis 200 mg/KgBB, kelompok AlCl3 200 mg/KgBB dengan Resveratrol 5 mg/KgBB dan kelompok AlCl3 200 mg/KgBB dengan Resveratrol 10 mg/KgBB. Hasil penelitian menunjukkan bahwa resveratrol berpengaruh secara signifikan pada mencit yang diinduksi aluminium klorida. Resveratrol dengan dosis 10 mg/KgBB merupakan dosis terbaik dalam menurunkan jumlah rerata mikronukleus yang terdapat pada mencit swiss webster. The accumulation of aluminum in the form of Al3+ ions in the body which is commonly found in daily life with a period of 7 years can cause toxicity. This is because the Al3+ ion has a strong affinity compared to iron (Fe) to bind to transferrin, thereby inhibiting the binding of oxygen in the blood and causing the production of Reactive Oxygen Species (ROS). This gives rise to DNA fragments which are indicated by the presence of a micronucleus. In reducing the occurrence of excess ROS production, natural ingredients are needed which contain stilbenoid groups that bind to oxidant compounds and free radicals, namely resveratrol. In Indonesia, resveratrol has been isolated from tempeh, but in vivo research is needed on its anti-genotoxic effect. The research method used was a completely randomized design with 4 treatments, namely the control group, the AlCl3 group at a dose of 200 mg/KgBW, AlCl3 200 mg/KgBW with Resveratrol 5 mg/KgBW and AlCl3 200 mg/KgBW with Resveratrol 10 mg/KgBW. The results showed that resveratrol had a significant effect on mice induced by aluminum chloride. Resveratrol at a dose of 10 mg/KgBW is the best dose in reducing the average number of micronuclei found in Swiss Webster mice
