Veterinary Medicinal Products in Manure: Development of a Technical Protocol for Laboratory Tests

Ziel der vorliegenden Arbeit war die Entwicklung eines Methodenkatalogs mit Richtliniencharakter zur Durchführung von Labortests zum Rückstandsverhalten von Tierarzneimitteln in Rinder- und Schweinegülle. Gülle stellt eine heterogene Probenmatrix hoher Komplexität dar, deren Zusammensetzung in Abhängigkeit von diversen Faktoren variiert. Die Entnahme einer Gülle mit reproduzierbarer Zusammensetzung aus landwirtschaftlichen Betrieben ist damit nicht möglich. Zur Gewinnung von Gülleproben für Labortests wurden gezielt Exkremente von Einzeltieren, welche unter definierten Bedingungen gehalten wurden, entnommen. Hieraus wurde Referenzgülle mit einem Trockensubstanzgehalt von 10 % (Rind) bzw. 5 % (Schwein) hergestellt. Es erfolgte eine Matrixcharakterisierung anhand folgender Parameter: Trockensubstanz (TS), mineralischer Gehalt (Rmin), Phosphor-Gehalt (P), organischer Gesamt-Kohlenstoff (TOC), pH-Wert, Redox-Potential (Eh), gelöster Sauerstoff (O2), Ammonium-Stickstoff (NH4-N), Gesamt-Stickstoff (Ntotal), biologischer Sauerstoffbedarf (BSB5). Der Einsatz der Referenzgülle zur Untersuchung der Stabilität von Tierarzneimitteln wurde anhand der 14C-markierten Antibiotika Sulfamethoxazol, dessen Hauptmetabolit Acetyl-Sulfamethoxazol, sowie das Strukturanalogon Sulfadiazin untersucht. Die Inkubation erfolgte in geschlossenen Laborbatchsystemen bei 20 °C im Dunkeln für 0, 3, 7, 30, 72, 100 und 177 Tage unter anaeroben Milieubedingungen. Aufgrund der erzielten Ergebnisse kann davon ausgegangen werden, dass bei Anwendung des beschriebenen Methodenkataloges eine repräsentative und reproduzierbare Referenzgülle für den Einsatz in Labortests hergestellt werden kann. Damit ist die reproduzierbare Durchführung eines Stabilitätstests von Tierarzneimitteln unter anaeroben Bedingungen möglich. Der hier vorgestellte Methodenkatalog stellt damit die Grundlage für eine Diskussion im Hinblick auf die Erstellung einer Richtlinie zum Test von Tierarzneimitteln in Gülle dar.This work has been targeted on development of a technical protocol for laboratory testing on the stability of veterinary medicinal products in liquid bovine and swine manures. Liquid manures are heterogeneous matrices of high complexity. During the storage up to several months, organic constituents and contaminants of manures underlie decomposition processes. Furthermore, residues of feeding stuff and straw as well as disinfection and cleaning agents may be released into the storage tanks by farming practices. Due to these aspects, the representative and reproducible sampling from tanks by farming practices is considered impossible. There¬fore, excrements of conventionally fed single cows and pigs were taken and then adjusted to dry matter contents of 10 % and 5 %, respectively, to reproducibly prepare manure. The characterization of the excrement and manure samples under study was based on the following parameters: dry substance (ds), mineral content (Rmin), phosphate content (P), total organic carbon (TOC), pH value, redox potential (Eh), dissolved oxygen (O2), ammonium nitrogen (NH4-N), total nitrogen (Ntotal), biological oxygen demand (BOD5). The experimental design of the stability tests have to be focused on practical relevance of agricultural and analytical processing. For these purposes the antibiotics sulfamethoxazole, the metabolite acetyl-sulfamethoxazole and sulfadiazine were used as 14C-labelled radiotracers to set up detailed mass balances. The incubation is conducted in the dark at 20 °C for 0, 1, 3, 7, 30, 72, 100 and 177 days under anaerobic conditions. Due to the results it is assumed that the use of representative and reproducible manure for laboratory testing on the stability of veterinary medicinal products under anaerobic conditions is possible.
