Terpenoids are a vast family of natural products utilised by humans as insect repellents, medicines and fragrances. The sesquiterpenoid (15-carbon terpenoid) artemisinin, a secondary metabolite of Artemisia annua, is the key antimalarial used to treat the devastating illness of malaria which kills around three million people a year. It is naturally generated by oxidative metabolism of the sesquiterpene amorpha-4,11-diene, produced from farnesyl diphosphate (FDP) by amorpha-4,11-diene synthase (ADS). Currently artemisinin is predominantly sourced by extraction from A. annua, which is converted from FDP in six steps, but more stable and cost-effective alternative sources are still desired.
