(알츠하이머 병에서 감마 시크리테이즈 조절 인자 발굴 및 기전 규명에 관한 연구)

학위논문(석사) -- 서울대학교대학원 : 자연과학대학 생명과학부, 2022.2. 정용근.Alzheimer's disease (AD) is a neurodegenerative disease that accounts for the largest proportion among the diseases in senior population, and AD patients shows pathological memory disorder known as dementia. There are several known genetic and environmental factors that are critical for AD pathogenesis, but the relevance between risk factors and molecular mechanism needs to be elucidated. Amyloid-β peptide (Aβ) is a main component of amyloid plaque, a pathologic hallmark of AD. Production and accumulation of Aβ plays a key role in the onset and progression of AD due to its neurotoxic oligomeric forms, and it is called ‘Aβ cascade hypothesis’. It has been known that γ-secretase is a membrane protease complex responsible for rate-limiting step of APP processing and generates Aβ40 and 42 after APP cleavage by BACE. Despite numerous efforts to find the exact mechanism of γ-secretase regulation, an important issue on how γ-secretase assembly is regulated still unclear. In this study, to isolate a novel regulator of γ-secretase activity, I screened about 1,806 Homo sapiens secretion proteins in mammalian expression vector with gain-of-function cell-based assay employing Tet-on/off system. By measuring C99-cleavage-induced GFP signal in the co-transfected cells, 20 positive candidate clones were isolated and confirmed for their effect on γ-secretase activity with the luciferase reporter assay. Among the putative positive clones, N12 (NUDT12) was the most effective to stimulate γ-secretase activity compared to control one. Interestingly, I found that over-expression of N12 increased formation of γ-secretase complex in BN-PAGE assay, while it did not increase levels of γ-secretase complex components in SDS-PAGE assay. Given that N12 has an activity modulating NAD, I found that this stimulatory effect of N12 was diminished by NAD supplementation in cell system. Since NAD is a major co-factor in cell metabolism, I further assessed upstream and downstream signals of N12 by employing serum-deprivation condition and PPARγ inhibitor, and found that the stimulating effect of N12 was abolished by serum-deprivation but not affected by PPARγ inhibitor, supporting that the increased level of γ-secretase complex by N12 overexpression is associated with NAD level. N12 knockdown did not affect activity of γ-secretase complex. Interestingly, while N12 overexpression increased AICD generation and stimulated amyloidogenic pathway, it did not affect Notch1 processing, showing APP-selective, Notch-sparing activity. Taken together, these results suggest that N12 positively regulates γ-secretase activity and increases level of γ-secretase complex by affecting cellular NAD level.알츠하이머 병(Alzheimer's disease, AD)은 노년층 인구에 대하서 가장 큰 비중을 차지하는 신경퇴행성 질환이며, 해당 환자들은 흔히 치매로 알려진 기억장애를 증상으로 나타낸다. 알츠하이머 병의 발병에 관여하는 유전인자 및 환경적 요인이 일부 밝혀졌지만, 외부 위험 요소와의 관련성과 분자적 기전을 보다 명확하게 규명할 필요가 있다. 알츠하이머병의 병리학적 특징인 노인반(Aβ plaque)을 구성하는 펩타이드인 아밀로이드-베타 펩타이드(Aβ)는 신경 독성을 나타내는데, 때문에 이의 생성과 축적이 알츠하이머 병의 발병과 진행에 중요한 역할을 맡고 있다는 'Aβ 연쇄 가설'을 기반으로 많은 병리학적 연구가 이루어진다. 감마 시크리테이즈(γ-Secretase)는 아밀로이드 전구 단백질을 절단하는 데에 있어 속도 제한 단계를 담당하는 막 단백질 분해 효소 복합체이다. 감마 시크리테이즈 활성의 정확한 조절 기전을 찾기 위해 많은 연구가 진행되었음에도 해당 영역에서는 여전히 많은 질문이 남아있다. 본 연구에서는 감마 시크리테이즈 활성에 관여하는 새로운 조절인자를 동정하기 위해 Tet-on/off 시스템을 사용하여 1,806개의 인간 세포 분비 단백질에 대한 스크리닝을 진행했다. HEK293T 세포에서 아밀로이드 전구 단백질의 C99 절단에 의해 유도된 형광 신호를 측정함으로써, 20개의 양성 후보 유전자를 분리한 후, 추가적으로 루시퍼레이즈 리포터 신호를 분석하여 감마 시크리테이즈 활성에 대한 효과를 확인했다. 그 중 N12 (NUDT12)는 대조군에 비해 감마 시크리테이즈의 활성도를 증가시키는 데에 효과적인 결과를 나타냈다. NUDT12의 과발현이 BN-PAGE 분석에서 감마 시크리테이즈 복합체의 형성을 증가시키는 반면, SDS-PAGE 분석에서 감마 시크리테이즈 복합체의 구성 단백질 각각의 양은 증가시키지 않는다는 결과를 확인했다. N12가 NAD를 조절하는 활성을 갖는다는 점을 고려하여 추가적인 실험을 진행했을 때, N12 과발현이 나타내는 효과가 NAD를 추가적인 처리에 의해 감소된다는 것을 확인했다. NAD는 세포 대사의 주요 보조 인자이며, 이의 조절 기전 경로에 있는 조건들을 달리하여 효과가 나타나는 지 비교했다. 세포 배양 시에 사용되는 혈청 농도의 결핍 조건과 PPARγ 억제제 처리 조건에서 N12의 효과가 복구되는 현상을 확인할 수 있었다. N12의 발현을 감소시킨 경우에는 감마 시크리테이즈 복합체의 양 및 활성에 영향을 미치지 않았지만, N12 과발현이 AICD 생성을 증가시키고 알파 및 베타 시크리테이즈 활성 역시 조절하여 아밀로이드 베타의 생성을 유도하는 과정을 촉진시키는 결과를 확인했다. 한편 감마 시크리테이즈의 또 다른 기질 단백질인 Notch1 절단에는 관여하지 않는 결과를 확인함으로써 감마 시크리테이즈가 아밀로이드 전구 단백질만을 분해하는 선택적인 활성 조절을 나타낸다고 추측할 수 있다. 종합하면, 본 연구에서는 N12가 감마 시크리테이즈의 양 및 활성을 조절하고, 이는 세포 내부의 NAD 양에 영향을 미침으로써 나타나는 현상임을 시사한다.ABSTRACT i CONTENTS iv LIST OF FIGURES v ABBREVIATION vii INTORDUCTION １ MATERIALS AND METHODS ５ RESULTS １０ DISCUSSION ４５ REFERENCES ４９ 국문 초록 ５７석
