Prebiotic chemistry : synthesis and reactivity of sulfur amino acids at the origin of life

La synthèse des acides aminés et des peptides est un des problèmes majeurs de la chimie prébiotique.Notre travail débute par une étude de la synthèse de la cystéine à partir du mercaptoacétaldéhyde. Le mercaptoacétaldéhyde est formé à partir du chloroacétaldéhyde qui lui-même est obtenu par chloration de l’acétaldéhyde. Nous avons ensuite cyclisé l’homocystéine dans l’eau pour former sa thiolactone, un cycle à 5 chaînons, capable de réagir avec des acides aminés et former des liaisons peptidiques. Nous avons testé la formation de peptides à partir la thiolactone de l’homocystéine ainsi qu’à partir de ses dérivés formylé et acétylé. Enfin, nous avons tenté de synthétiser la S-méthyl méthionine, SMM, un donneur de méthyle potentiel, analogue simplifié de la S-adénosyl méthionine, SAM. Un intermédiaire important a été obtenu, mais notre voie de synthèse n’a pas conduit à la SMM.
