Moringa oleifera (Moringaceae) and Lepidium meyenii (Brassicaceae) as superfood : cytotoxic effect of given extracts

Cilj istraživanja: Cilj istraživanja je ispitati potencijalno citotoksično djelovanje biljnih vrsta Moringa oleifera i Lepidum meyenii na humane karcinomske stanice dojke (MDA-MB-231) i mokraćnog mjehura (T24). Pretpostavka je da će se broj karcinomskih stanica nakon izlaganja uzorcima smanjiti u odnosu na kontrolnu skupinu. Materijali i metode: Ispitivanje citotoksičnosti na staničnim linijama karcinoma dojke i mokraćnog mjehura rađeno je MTT metodom kojom se određuje postotak metabolički aktivnih stanica nakon tretiranja ekstraktima dobivenima autolizom. Uzorci biljaka pripravljeni su u pet različitih koncentracija (1 µg/mL, 5 µg/mL, 10 µg/mL, 50 µg/mL i 100 µg/mL), a citotoksični učinak se određivao nakon 4, 24, 48 i 72 sata inkubacije. Djelotvornost ekstrakata određena je spektrofotometrijski mjerenjem apsorbancije pri 570 nm. Rezultati: Rezultati su prikazani grafički u odnosu vremena inkubacije i postotka metabolički aktivnih stanica. Kod stanica karcinoma mokraćnog mjehura (T24) svi uzorci ostvaruju bolji učinak pri višim koncentracijama od 50 µg/mL i 100 µg/mL. Uzorak crvene mace pokazuje najznačajniji učinak na smanjenje metabolički aktivnih stanica nakon 72 h inkubacije pri koncentraciji od 50 µg/mL. Također, zabilježena je IC50 vrijednost od 17,57 µg/mL što se karakterizira kao značajno aktivan ekstrakt. Kod stanica karcinoma dojke, svi uzorci pokazali su umjerenu citotoksičnu aktivnost pri višim koncentracijama od 50 µg/mL i 100 µg/mL. Najznačajniji učinak pokazuje uzorak biljke Moringa oleifera pri koncentraciji od 50 µg/mL nakon 72 h inkubacije. Zaključci: In vitro izlaganje stanica karcinoma dojke (MDA-MB-231) i mokraćnog mjehura (T24) ekstraktima dobivenim iz biljnih vrsta Moringa oleifera i Lepidium meyenii dovodi do smanjenja preživljenja tih stanica. Citotoksični učinak ovisan je o vremenu inkubacije i koncentraciji. Citotoksični učinak biljnih vrsta Moringa oleifera i Lepidium meyenii, ujedno i hipoteza našeg ispitivanja, je potvrđen. Potvrđene učinke potrebno je ispitati in vivo na životinjskim modelima.The aim of the research: The aim of the research is to examine the potential cytotoxic effects of Moringa oleifera and Lepidium meyenii plants on human carcinoma cells of breast (MDA-MB-231) and bladder (T24). The assumption is that number of carcinoma cells after the exposure to plant extracts will be reduced in comparison to the control group. Material and methods: The cytotoxicity was tested by the MTT method used to determine the percentage of metabolically active cells after exposure to plant extracts obtained by autolysis. Plant samples were prepared at five different concentrations (1 µg/mL, 5 µg/mL, 10 µg/mL, 50 µg/mL and 100 µg/mL), and the effect is seen after 4, 24, 48 and 72 hours. The efficacy of the extracts was observed spectrophotometrically after measurement of absorbance at 570 nm Results: The results are graphically presented in relation to incubation times and the percentages of metabolically active cells. In bladder carcinoma cells all of the samples show better effects in higher concentrations of 50 µg/mL and 100 µg/mL. The most prominent effect on the reduction of metabolically active cells is achieved by red Lepidium meyenii sample after 24 hour incubation time in a concentration of 50 µg/mL. Also, IC50 value of 17,57 µg/mL was detected, which is characteritzed as a very active extract. In breast cancer cells, all of the samples showed moderate cytotoxic activity at the higher concentrations of 50 µg/mL and 100 µg/mL. The most significant effect was achieved by the sample of Moringa oleifera at a concentration of 50 µg/mL after 72-hour incubation time. Conclusions: In vitro exposure of human carcinoma cells of breast (MDA-MB-231) and bladder (T54) to extracts obtained from Moringa oleifera and Lepidium meyenii leads to reduced cell survival rate. Cytotoxic effect depends on incubation time and concentration. The cytotoxic effect of Moringa oleifera and Lepidium meyenii plants, which was the hypothesis of the research, is confirmed.
