Extract and molecules from Aloe arborescens leaf tested on murine

Questa Tesi ha lo scopo di contribuire ad ampliare le conoscenze scientifiche sulle proprietà biologiche della pianta Aloe arborescens. Saggi MTT hanno mostrato un'intensa attività antiproliferativa dell’estratto di epidermide fogliare di Aloe arborescens testato su cellule P3X murine e HL60 umane. Studi citologici sono stati effettuati sia con tecniche di immunofluorescenza, sia mediante osservazioni SEM e TEM. Analisi chimiche sono state effettuate mediante tecniche TLC, HPLC e LC-Massa che hanno mostrato la presenza nell’estratto di quattro molecole principali: aloenina, aloina A, aloina B e 2’O-feruloylaloesina. L’analisi NMR ha mostrato nello spot E, isolato mediante TLC e risultato biologicamente attivo, la presenza delle prime tre molecole. Singoli standard molecolari di aloenina, aloina A e B e soluzioni formate dalla diversa combinazione di esse sono stati testati su cellule P3X e HL60. Nella somministrazione di estratti foglia di Aloe arborescens a basse concentrazioni per lunghi periodi alcune cellule di mieloma murino erano sopravvissute, l'attenzione si è concentrata alle cellule superstiti e l'induzione di resistenza di farmaco-correlato, queste cellule hanno mostrato un aumento del proprio volume, continuando a proliferare. Al fine di valutare il meccanismo molecolare coinvolto, è stata testata l'espressione delle proteine PGP e p53 mediante tecniche di western blot e indagini di immunofluorescenza. Inoltre cellule di controllo non trattate con estratto di Aloe sono state fatte crescere in terreno di coltura di cellule resistenti e hanno mostrato di acquisire proprietà di resistenza, confermando così la presenza nel terreno di coltura di un fattore di induzione di resistenza ai farmaci.This thesis was carried out in order to contribute to enlarge knowledge on the biological properties of Aloe arborescens plant. MTT assays showed an intense antiproliferative activity of Aloe arborescens leaf skin extracts tested on murine myeloma cells (P3X) and human promyelocytic cells (HL60). Cytological study have been carried out with immunofluorescence techniques and with SEM and TEM observations. Chemical analysis of the leaf extract with TLC, HPLC and LC-mass investigations showed the presence of the molecules aloenin and the aloins A and B, 2’O-feruloylaloesin. The spot E, isolated by TLC, showing antiproliferative activity and NMR investigations showed that this spot consisted of the molecules aloenin and the aloins A and B. The concentration of single molecules was confirmed by HPLC investigations and single molecules and solutions formed by different combination of single molecules were tested on P3X and HL60. Since after administration of few concentration for long time of Aloe arborescens leaf extract some murine myeloma cells survived, attention was focussed to the surviving cells and related induction of drug-resistance properties; such cells showed an increasing of cell volume and were able to proliferate. In order to evaluate a possible molecular marker, the expression of PGP and p53 proteins ware tested by western blot techniques and immunofluorescence investigations. Further, control cells untreated with Aloe arborescens leaf extract were then cultured in culture medium of resistant cells and drug resistant properties resulted acquired, thus confirming the presence in the culture medium of a drug resistance inducing factor.Dottorato di ricerca in Genetica e biologia cellular
