PENGARUH SEDIAAN EKSTRAK KERING DAUN KELOR (Moringa oleifera Lmk.) TERHADAP PENURUNAN KADAR KOLESTEROL TOTAL DARAH TIKUS PUTIH JANTAN GALUR WISTAR

Hypercholesterolemia is a risk factor for stroke and coronary heart disease. This is due to the increased levels of LDL (Low Density Lipoprotein) which caused buildup of blood on vessel walls and form plaques that cause ateroschelosys, which is hardening and narrowing of blood vessels. Moringa (Moringa oleifera Lmk.) leaves is a plant that can be used to lower the cholesterol levels. This study aimed to determine the effects of Moringa leaves that made in the preparation of the dry extract to the decrease of total cholesterol levels of male mice fed a high-fat diet and to determine the effective dose. The rats were divided into 6 groups, each consisting of 5 rats. Measurement of total cholesterol levels on day 0, day 14, and day 28. The hypercholesterolemia conditions done by feeding a high fat diet that is lard and yolk for 14 days. The dry extract preparation test given after the mice had hypercholesterolemia. The dose of Moringa leaf dry extract preparation on mice was given a dose I: 32 mg/200 g BW, dose II: 63 mg/200 g BW, and dose III: 95 mg/200 g BW. The positive control used was simvastatin and CMC negative controls of 0.5%, while for untreated normal controls, only fed BR II alone. Test preparation is administered for 14 days, and then the data were analyzed by one way ANOVA. The results showed the decrease in total cholesterol levels in all three dose variation of Moringa leaf dry extract dosage on day 28. The dry extracts preparations dose of 32 mg/200 g BW showed cholesterol-lowering equal to the effect of simvastatin.   Keywords: dry extract, moringa leaves, total cholesterol level, simvastatin.Hiperkolesterolemia merupakan salah satu faktor penyebab stroke dan jantung koroner. Hal ini disebabkan karena terjadinya peningkatan kadar LDL (Low Density Lipoprotein) yang menyebabkan terjadinya penumpukan pada dinding pembuluh darah  dan membentuk plak sehingga menyebabkkan ateroskelosis, yaitu pengerasan dan penyempitan pembuluh darah. Daun kelor (Moringa oleifera Lmk.) merupakan tanaman yang dapat digunakan untuk menurunkan kadar kolesterol. Penelitian ini bertujuan untuk mengetahui efek daun kelor yang dibuat dalam sediaan ekstrak kering terhadap penurunan kadar kolesterol total tikus putih jantan yang diberi diet tinggi lemak dan untuk mengetahui dosis efektifnya. Tikus-tikus dibagi menjadi 6 kelompok masing-masing terdiri dari 5 ekor tikus. Pengukuran kadar kolesterol total dilakukan pada hari ke 0, hari ke 14, dan hari ke 28. Kondisi hiperkolesterolemia dilakukan dengan pemberian pakan diet tinggi lemak yaitu lemak babi dan kuning telur selama 14 hari. Sediaan uji ekstrak kering diberikan setelah tikus mengalami hiperkolesterolemia. Dosis sediaan ekstrak kering daun kelor yang diberikan pada tikus adalah dosis I : 32 mg/200 g BB, dosis II : 63 mg/200 g BB, dan dosis III : 95 mg/200 g BB. Kontrol positif yang dipakai adalah simvastatin dan kontrol negatif  CMC 0,5 %, sedangkan untuk kontrol normal tidak diberi perlakuan, hanya diberi pakan BR II saja. Sediaan uji diberikan selama 14 hari, kemudian data yang diperoleh dianalisis dengan ANOVA satu jalan. Hasil penelitian menunjukkan adanya penurunan kadar kolesterol total pada ketiga variasi dosis sediaan ekstrak kering daun kelor pada hari ke 28. Sediaan esktrak kering dosis 32 mg/200 g BB menunjukkan efek penurunan kadar kolesterol yang setara dengan efek simvastatin.   Kata kunci : Ekstrak kering, daun kelor, kadar kolesterol total, simvastatin
