산뽕나무로부터 분리한 mulberrofuran G의 산소-포도당 결핍 유도성 세포 사멸에 대한 보호 효과 : NADPH oxidase, ROS생성 및 ASK1 인산화의 억제

학위논문 (석사)-- 서울대학교 대학원 : 약학과, 2012. 2. 마응천.The pathogenesis of cerebral ischemic injury which is characterized by reduction of blood supply and oxygen and nutrients deprivation in brain is very complicated. High-potential pathway is implicated with reactive oxygen species (ROS) overproduction and NADPH oxidase (Nox) system is one of the main source of ROS which leads to consequent oxidative stress in development of cerebral ischemia. In the recent studies, cerebral ischemia causes severe impairment of endoplasmic reticulum (ER) function which is called ER stress. NADPH oxidase has been revealed to link oxidative and ER stress and triggers apoptotic cell death pathway in neurodegenerative diesases. ROS-induced oxidative stress and ER stress induced by ischemic damage are intergrated into ASK1 which switch on apoptosis and neuronal cell death. In the present study, oxygen and glucose deprivation and reoxygenation (OGD/Reoxygenation) induced intracellular NADPH oxidase activity with Nox mRNA level increase and ROS overproduction in human neuroblastoma SH-SY5Y cells. In this study, mulberrofuran G, a diels-alder type adduct which was isolated from Morus bombycis (M. bombycis), protected neuronal cells against OGD/Reoxygenation by reducing NADPH oxidase activity with Nox1, 2 and 4 mRNA levels and ROS production. In addition, mulberrofuran G markedly suppressed OGD/Reoxygenation-induced spliced X-box1 (sXbp1), an ER stress biomarker, mRNA level. In western blot analysis, OGD/Reoxygenation-induced phosphorylation of ASK1 protein level was decreased by non-toxic dose range of mulberrofuran G treatment. Taken together, this present study indicated that the protective effect of mulberrofuran G against OGD/Reoxygenation-induced cell damage might be implicated with potent inhibition of Nox-mediated oxidative stress and, at least in part, ER stress-associated ASK1 protein activation in in vitro cerebral ischemic injury.뇌 허혈은 뇌혈관이 협착 되거나 혈전이 막히게 되면 혈액 공급이 차단되어, 산소와 영양분의 결핍이 일어남으로써 일어나게 된다. 일반적으로 뇌 허혈의 70~80%는 혈전의 막힘으로 인한 혈액공급의 일시적 중단과 순간적인 혈전 제거에 따른 급격한 혈액 재공급으로 인하여 뇌 기능의 부분적, 전체적 손상을 일으킨다고 보고되어 있다. 본 연구에서는 human neuroblastoma SH-SY5Y cells에 실제 임상학적 뇌 허혈을 유발하는 모델인 산소-포도당 결핍 (oxygen-glucose deprivation)을 통한 세포 사멸을 유도하여 혈액의 공급 중단에 따른 허혈 상태와 유사한 환경을 설정해 주었다. 이러한 전제 하에 산뽕나무에서 분리된 단일물질인 mulberrofuran G이 산소-포도당 결핍이 유도된 SH-SY5Y cells과 흰 쥐의 뇌 조직 절편에서 mulberrofuran G가 세포 및 뇌 절편의 생존률(%)을 농도 구배적으로 증가시키는 뇌 신경세포의 보호 효과가 있는 것을 확인하고 그 구체적인 작용 기전을 규명하고자 하였다. NADPH oxidase (Nox)는 주요한 활성 산소종 생성계로, 과다한 활성 산소종(reactive oxygen species, ROS)의 생성은 산화적 스트레스를 초래하며 염증반응, 세포 고사 등을 유발하여 세포 손상을 야기한다. 특히 최근 연구들에서 Nox는 in vitro 및 in vivo cerebral ischemia에서 활성 산소종을 생성하고, 흥분성 신경세포독성과 함께 염증 반응을 유발하여 뇌허혈에 의한 세포 손상 및 사멸을 일으키는 주요한 요인으로 보고되고 이와 함께 Nox가 소포체 및 산화적 스트레스, 세포고사 기전의 매개 인자라고 밝혀졌다. 본 연구에서 mulberrofuran G의 Nox에 대한 활성을 실험한 결과, 정상 세포에 비해 산소-포도당 결핍된 세포에서 47% 가량 증가한 NADPH oxidase 효소 활성을 2 μM의 mulberrofuran G 처리 시 32% 줄이는 것을 확인하였다. 또한, 산소-포도당 결핍에 의해 Nox1, Nox2 및 Nox4 mRNA 발현은 각각 정상 세포의 1.4, 1.5 및 2.9배 증가했으며, 2 μM의 mulberrofuran G는 각 mRNA 발현을 정상 세포의 1.0, 1.0 및 1.5배로 감소시켰다. 이와 함께, 산소-포도당 결핍에 의해 38% 증가한 ROS발생을 2 μM의 mulberrofuran G 가 19% 감소시켰다. 소포체 스트레스의 지표 인자로서 X-bot protein 1(Xbp1)은 뇌 허혈 등에 의한 소포체 스트레스 신호에 의해 mRNA의 splicing이 일어난다고 알려졌는데, 본 연구에서 산소-포도당 결핍에 의해 SH-SY5Y cell의 spliced Xbp1 mRNA 발현이 2.3배 증가하였고 2 μM의 mulberrofuran G가 이를 0.9배로 감소시켰다. ASK1은 ROS 및 소포체 스트레스에 의해 인산화되어 활성화되는 세포 자멸사 유도 인자로 알려져있는 단백질이며, 본 연구에서 산소-포도당 결핍에 의해 ASK1 단백질이 2.4배 활성화 되었고, mulberrofuran G의 처리가 이를 농도 구배적으로 감소시킴을 확인하였다. 이러한 결과들을 바탕으로, mulberrofuran G가 산소-포도당 결핍에 의해 활성화된 NADPH oxidase의 효소 활성 및 구체적으로 Nox1, Nox2 및 Nox4 mRNA의 과발현을 억제하고, 활성 산소종의 과생산을 억제함으로써 최종적으로 소포체 스트레스 연관 세포 자멸사를 일으키는 주요한 단백질인 ASK1을 억제하고, 이와 함께 소포체 스트레스 기전의 주요 매개 인자인 Xbp1의 splicing을 억제하였으며, 이를 통해 mulberrofuran G가 산소-포동당 결핍에 의해 유도된 신경 세포 독성과 사멸에 대한 보호 효과가 있음을 규명하였다.Maste
