ESTUDO COMPARATIVO DO EFEITO DA AMBIENTAÇÃO E DOS ANTICOAGULANTES EDTA E CITRATO DE SÓDIO SOBRE A AGREGAÇÃO PLAQUETÁRIA EM AMOSTRAS SANGUÍNEAS DE GATOS DOMÉSTICOS

The purpose of the research was to analise the aclimatation and two anticoagulants effects on platelet aggregation and compare the manual and automatic platelet counting methods in blood samples of cats. One group of ten animals was submitted to a pre blood collection aclimatation under the effects of a facial pheromone difusor, one hour long. The other group of tem cats either, did was not submitted to the pre-collection aclimatation. In both groups was collected 1 mL of blood from the saphenous vein with a n.23 scalp and 3 mL syringe. Half volume was transfered to an eppendorf container with EDTA and the other half was transfered to an eppendorf container with sodium citrate 3.2%. The samples was processed on the Clinical Pathology Laboratory of the UFPB Veterinary Hospital, by automatic counter and then the blood smears was prepared. The platelet aggregations was subjectively classified according to the amount of clumps on the lateral margins and feathered edge as discreet, moderate and intense. Only the samples with discreet and moderate aggregation was submitted to the manual counting in Neubauer chamber and blood smear estimation. The results shown that there was no significant statistical difference in the platelet counts between the aclimated and the non aclimated groups. There was significant difference between the blood smear counting and the automatic counting and between the anticoagulants EDTA and sodium citrate 3.2%. The sodium citrate 3.8% was largely responsible for the intense platelet aggregations.O objetivo do trabalho foi analisar o efeito da ambientação e de dois anticoagulantes sobre a agregação plaquetária, bem como comparar os métodos de contagem manual e automática de plaquetas em amostras sanguíneas de gatos. Um grupo, formado por dez animais, foi submetido à uma hora de ambientação com a sala difundida por feromônio facial felino sintético, antes da colheita de sangue. O outro grupo, composto por dez gatos, não passou pelo período de ambientação pré-colheita. Em ambos os grupos, colheu-se 1ml de sangue da veia safena medial, com auxílio de scalp n.23 acoplado à seringa de 3ml. Metade do volume foi depositado em tudo de polipropileno com EDTA e metade em tubo de polipropileno com citrato de sódio a 3,2%. No Laboratório de Patologia Clínica do Hospital Veterinário da UFPB, as amostras foram processadas em aparelho automático e os esfregaços sanguíneos confeccionados. As agregações plaquetárias foram classificadas subjetivamente, de acordo com a quantidade de grumos presentes nas bordas e na franja do esfregaço em: discretas, moderadas e intensas. Somente as amostras com nível de agregação discreto ou moderado, foram submetidas à contagem estimada das plaquetas em câmara de Neubauer e no esfregaço sanguíneo. Os resultados mostraram que não houve diferença estatística entre as contagens de plaquetas dos gatos ambientados comparativamente aos não ambientados. Houve diferença estatística entre a contagem manual estimada em lâmina e a contagem automática e também entre os anticoagulantes EDTA e citrato de sódio a 3,8%, sendo o último responsável pela maior parte das agregações plaquetárias intensas
