Investigation of cytogenetic effects of some pesticides on different plant genotypes

Bu çalışmada Nuarimol, Fenarimol ve Previcur-n (propamocarb hydrochloride) olarak adlandırılan fungisidlerin, Allium cepa L. ev kantar topu, Allium cepa L. ev. imralı, Vida foba ev. fîliz 99 ve Vida foba ev. ereksen 87 olarak bilinen çeşitlerdeki sitogenetik etkilerinin araştırılmasına çalışıldı. Bu üç fungisid çeşidi, çeşitli dozlarda (25, 50, 100, 100, 600 ve 25, 50, 100, 200, 400 mg./mL) ve önceden belirlenen sürelerde (2, 4, 6, 8, 16, 24 ve 3, 6, 12, 24 saat) muamele edilerek, kromozomlar üzerindeki etkilerine bakıldı. Bu amaçla elde edilen kök uçları ezme preperat yöntemi kullanılarak ve asetoorsein boyası ile boyanarak hazır preperat haline getirildi. Hazırlanan preperatların her birinde 1000 hücre 4 tekrarlı olarak sayımı yapılarak incelendi. Yapılan çalışma sonuçlarına göre; doz ve uygulama sürelerine bağlı olarak her üç fungisid çeşidi de mitotik indexin azalmasına neden olmuştur. Bunun yanında anafaz köprüleri, c-metafaz figürleri, kromozom kırıklıklarının doz artışına ve uygulama sürelerine bağlı olarak arttığı gözlendi.In this study, the cytogenetic effects of some fungicides named Ptevicur-n (propamocarb hydrochloride), Fenarimol and Nuarimol were studied on Allium cepa L. cv. kantartopu, Atliwn cepa L. cv. imrah, Vida faba ev. filiz 99 and Viciafaha cv. ereksen 87. These three fungicides were treated with varying doses of 25, 50, 100, 300, 600 and 25, 50, 100, 200, 400 mg./mL. and duration of 2, 4, 6, 8, 16, 24 and 3, 6, 12, 24 hour to examine the effects on the chromosomes of Allium cepa L. cv. kantartopu, Allium cepa L. cv. imrali, Viciafaha cv. fifiz 99 and Viciafaha cv. ereksen 87. For this study, root tips were prepared by using squash method and stained with acetoorcein. From each slides, T000 cells were examined four times. According to results, all the fungicides (Previcur-n, Fenarimol and Nuarimol) reduced the mitotic index. At the same time, chromosome bridge, c-methaphase figures and chromosome breaks increased dose and treatment time dependently
