Examining the apoptotic effect of resveratrol on MCF-7 cell strain

Gereç ve yöntemler: Çalışmalarımızda, hücrelere verilecek olan resveratrol dozları 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromid (MTT) analizi ile belirlendi. Resveratrolün MCF-7 hücre soyundaki apoptotik etkisi, APC anneksin V antikoru kullanarak akım sitometri görüntüleme yöntemi ile belirlendi. Bulgular: 100 μM ve 250 μM resveratrol MCF-7 hücrelerine inkübe edildi ve ölçümler 12, 24. ve 48. saatlerde elde edildi. MTT sonuçlarından, resveratrolün artan konsantrasyonu ile doğru orantılı olarak hücre yoğunluğunun kontrol grubuna göre azaldığı tespit edildi. On ikinci saatte ölçülen analizlerde sitotoksik doz 250-500 μM arasında, 24. saatteki ölçümlerde 100-250 μM arasında, 48. saatteki ölçümlerde ise 100 μM olarak belirlendi. Anneksin V akım sitometri ölçüm sonuçları değerlendirildiğinde; MCF-7 hücrelerine verilen 100 ve 250 μM resveratrolün kontrol grubuna kıyasla erken apoptozu artırdığı gözlemlendi. Sonuç: Resveratrolün tüm bu antikanser etkilerinden biri olan apoptotik etkisi hem doz hem de zamana bağlı olarak MCF-7 meme kanseri hücre soyunda özellikle erken apoptozu indüklediği gözlemlenmiştirObjectives: This study aims to examine the cytotoxic effect of resveratrol, as a natural polyphenolic compound in vitro environment, in MCF-7 breast cancer cell strain. Materials and methods: We determined the doses of resveratrol with 3-(4.5-dimethylthiazol-2-yl)-2.5-diphenyltetrazolium bromide (MTT) analysis. We detected the apoptotic effect of resveratrol on MCF-7 cell strain by the imaging flow cytometry using the APC annexin V antibody. Results: 100 μM and 250 μM resveratrol were incubated in MCF-7 cells and measurements were obtained at the 12, 24 and 48 hours. From the MTT results, we found that the cell density decreased with respect to the control group, directly proportional to the increasing concentration of resveratrol. We determined the cytotoxic doses to be between 250-500 μM in the analysis of the 12th hour, between 100-250 μM at the 24 hour measurement and 100 μM at the 48 hour measurement. When the results of Annexin V flow cytometry measurements were evaluated; we observed that 100 and 250 μM resveratrol given to MCF-7 cells increased early apoptosis compared to the control group.
