Development of enzyme-linked immunosorbent assay method for diagnosis of infectious pancreatic necrosis disease

İnfeksiyöz pankreatik nekrozis (İPN), genç salmonid balıklarda yüksek mortalite ile seyreden ve önemli ekonomik kayıplara neden olan akut bulaşıcı viral bir hastalıktır. Bu çalışmada hastalığının teşhisinde standart bir metot olan enzim-linked immunosorbent assay (ELİSA) geliştirilmesi ve İPN hastalığının teşhisi için kullanılması amaçlanmıştır. İnfeksiyöz pankreatik nekrozis virusunun (İPNV) teşhisi hücre kültüründe izolasyonu takiben serolojik tekniklerle identifikasyon ile yapılmaktadır. Bunun için İPNV’unun VR-299, Ab ve Sp serotipleri pürifiye edilmiş ve bu serotiplere karşı tavşan ve kobaydan hiperimmun serum elde edilmiş ve bu antikorlar testte kullanılmıştır. Testin spesifitesini belirlemek için salmonid balıklarda hastalığa neden olan diğer viruslardan infeksiyöz hematopoetik nekrozis virus (İHNV) ve viral hemorajik septisemi virusu (VHSV) kullanılmış ve çapraz reaksiyon görülmemiştir. Testin sensitivitesi VR-299 serotipi için $DKID _{50}$ değeri $10^ {6.6}$/ml, Ab serotipi için $DKID _{50}$ değeri $10^ {6.8}$/ml ve Sp serotipi için $DKID _{50}$ değeri $10^ {7.3}$/ml olarak tespit edilmiştir. Sonuç olarak İPN hastalığının teşhisi için geliştirilen ELİSA yönteminin hızlı, basit ve ekonomik bir yöntem olarak kullanılabileceği düşüncesine varılmıştır.Infectious pancreatic necrosis (IPN) is an acute contagious disease and causes high mortality and significant economic losses in young salmonid fish. In this study, ELISA that is used as a standart method was developed to be used for diagnosis of IPN. The diagnosis of IPN virus is made by virus isolation followed by serological identification technics. For this purpose VR 299, Ab and Sp serotypes of infectious pancreatic necrosis virus was purified and then hiperimmun sera were produced in rabbit and guinea pigs against these serotypes. Finally these antibodies were used in the test. Other viruses like infectious hematopoietic necrosis virus and viral hemorrgagic septisemia virus that cause diseases in salmonid fish were used to determine the specificity of the test and cross reaction was not seen. Sensitivity of the test was found to be $10^ {6.6}$/ml, $10^ {6.8}$/ml, and $10^ {7.3}$/ml $TCID _{50}$ for Vr 299, Ab and Sp respectively. As a result, development of ELISA method was considered to be simple, rapid, and economic method to be used for diagnosis of ınfectious pancreatic necrosis disease
