Detection of infectious pancreatic necrosis virus (IPNV), SHSV (viral hemorrhagic septicemia virus),IHNV (infectious hematopoietic necrosis virus)in trout and virulence based on VP2, VP5 gene regions

Türkiye'de gökkuşağı alabalığı (Oncorhynchus mykiss) işletmelerinde İnkefsiyöz Pankreatik Nekroz Virüsü (IPNV) yaygın iken Viral Hemorajik Septisemi Virüsü (VHSV) 2006'da tespit edilmiş ancak İnfeksiyöz Hematopoietik Nekroz Virüs (IHNV) varlığı bildirilmemiştir. Bu çalışmada Türkiye'nin 6 farklı bölgesinde IPNV, VHSV, IHNV varlığı ve IPNV izolatlarının VP2 ve VP5 gen bölgelerinin filogenetik gruplandırması yapılarak virülensleri incelenmiştir. Virüs izolasyonu BF-2 ve EPC hücre kültürleri kullanılarak, identifikasyonu ise ELISA (Enzyme Linked Immuno Assay) ve RT-PCR (Ters Transkriptaz-Polimeraz Zincir Reaksiyonu) ile yapılmıştır. IPNV suşlarının genogrupları ve virülens motiflerini belirlemek için dizi analizi yapılmıştır. IPNV suşlarının immun gen uyarımlarının belirlenmesi amacıyla ise TO (Atlantik somon ön böbrek hücre hattı) hücre kültürü kullanılmış ve bu hücre kültüründeki IFN, Mx ve Viperin gen miktarları RT-qPCR (Eş Zamanlı Ters Transkriptaz-Polimeraz Zincir Reaksiyonu) ile ölçülmüştür. Çalışmada VHSV ve IHNV tespit edilemezken IPNV'nin tüm bölgelerde yaygın ve prevalansının %27,14 olduğu saptanmıştır. VP2 gen bölgesinin sekans ve filogenetik analiz sonuçları Türkiye'den izole edilen IPNV suşlarının genogrup 5'te (serotip Sp-A2) yer aldığını ve amino asit dizilimine göre tüm suşların P217T221 motifini barındırdığını göstermiştir. VP5 gen bölgesi nükleotit ve amino asit dizi analiz sonuçları suşların birbirlerine %97-100 benzer olduğunu ortaya koymuştur. VP2 ve VP5 gen bölgelerinin filogenetik analiz sonuçları ile suşların coğrafi kökenleri arasında herhangi bir ilişki saptanamamıştır. Hücre kültüründe yapılan immun yanıt çalışmalarında suşların IFN, Mx ve Viperin genlerini uyardığı tespit edilirken suşlar arasında immun yanıt uyarımlarının anlamlı sonuçlar vermediği tespit edilmiştir.Infectious Pancreatic Necrosis Virus (IPNV) in rainbow trout (Oncorhynchus mykiss) farms in Turkey is prevalent while in VHSV (Viral Hemorrhagic Septicemia Virus) has been reported in 2006, but IHNV (Infectious Hematopoietic Necrosis Virus) are detected yet. In this study, presence of IPNV, VHSV, IHNV and phylogenetic analysis of VP2 and VP5 genes and virulence of IPNV in six different regions of Turkey were examined. Virus isolation was performed using BF-2 and EPC cell cultures, and identification was performed by ELISA and RT-PCR. Sequence analysis was performed to determine the genogroups and virulence motifs of IPNV strains. TO cell culture was used in order to determine the induction of immune genes and IFN, Mx and Viperin genes was measured by RT-qPCR. In all of Prevalance of IPNV was found 27.14% while VHSV and IHNV were not detected. IPNV VP2 gene region was sequenced for analysis of phylogenetic examinationand IPNV starins of were belomged to genogroup 5 (serotype Sp-A2) and the amino acid sequence showed all strains had P217T221 amino acid motif. The VP5 gene region nucleotide and amino acid sequence analysis results showed that the strains were 97-100% similar to each other. The phylogenetic analysis results of VP2 and VP5 gene regions did not reveal any relationship between the strains and their geographical origins. However IFN, Mx and Viperin immune genes were induced by IPNV stains, there were no significant results.Yüksek Öğretim Kurum
