Development of analytical methodologies based on mass spectrometry and infrared spectroscopy for the identification of organic binders and their degradation products in colonial art

Las investigaciones realizadas en este trabajo se centraron en el estudio de los procesos de degradación de triacilglicéridos y proteínas en aglutinantes como aceites vegetales, huevo y cola animal aplicados sobre portaobjetos de vidrio y en contacto con yeso empleado como soporte en la pintura mural andina. Se determinó además la influencia en estas reacciones de mezclas pigmentarias conteniendo índigo. Se trabajó con réplicas pictóricas envejecidas naturalmente en condiciones del laboratorio así como por envejecimiento acelerado por irradiación con luz UV, preparadas según manuales y tratados de la época sobre técnicas empleadas en la elaboración de pintura mural. La primera parte del trabajo comprendió el análisis de muestras extraídas de réplicas pictóricas por FTIR-ATR, mediante el análisis de las derivadas segundas de los espectros, lo cual permitió identificar bandas características de cada aglutinante y visualizar mejor los cambios producidos por el envejecimiento de las muestras. Los extractos lipídicos de las réplicas pictóricas conteniendo aceites vegetales y huevo se analizaron por CG-FID y CG-EM. Se identificaron ácidos dicarboxílicos originados en reacciones de polimerización y oxidación de ácidos grasos insaturados. La formación de estos productos de oxidación fue mayor en réplicas preparadas sobre soporte de vidrio, sugiriendo una mayor estabilidad de los aglutinantes aplicados sobre yeso como base de preparación. Los aglutinantes proteicos mostraron mayor estabilidad que los lipídicos y las composiciones en aminoácidos no presentaron mayores variaciones en función del soporte. Los resultados obtenidos por FTIR ATR y CG-FID fueron analizados aplicando modelos estadísticos multivariados, permitiendo una mejor interpretación de los datos. En la segunda parte del trabajo se presentan los resultados obtenidos en la caracterización de aglutinantes orgánicos en pinturas murales sobre yeso de dos iglesias del siglo XVIII del área andina aplicando las técnicas analíticas y las metodologías optimizadas con las muestras de referencias. Se identificó la presencia de aceite vegetal y huevo como aglutinante de los pigmentos, característico de una pintura al temple, y el uso de una cola animal como imprimación de la base de preparación de yeso. La identificación de proteínas características de huevo y colágeno de la cola animal fue confirmada por MALDI-TOF. Estos resultados adquirieron relevancia al contrastarse con información suministrada por historiadores y restauradores de arte, constituyendo el primer estudio químico sobre la técnica pictórica en pintura mural andina.The research carried out in this work is focused on the study of the degradation processes of triacylglycerides and proteins in binders such as vegetable oils, egg and animal glue, applied on glass slides and in contact with gypsum used as support in Andean mural paintings. We determined the influence of pigment mixtures containing indigo in these reactions. We worked with pictorial replicas either aged naturally in laboratory conditions or by accelerated aging by irradiation with UV light. Replicas were prepared according to manuals and treaties of the age on techniques used in the preparation of mural painting. The first part of the work included the analysis of samples taken from replicas, by FTIRATR, through the analysis of the second derivatives of the spectra. This allowed the identification of the characteristic bands of each binder and to better visualize the changes produced by aging the samples. The lipid extracts of the pictorial replicas containing vegetable oils and egg were analyzed by CG-FID and CG-EM. Dicarboxylic acids originated in polymerization and oxidation reactions of unsaturated fatty acids were identified. The formation of these oxidation products was higher in replicas prepared on glass support, suggesting a higher stability of the binders applied on gypsum as a base for preparation. The protein binders showed greater stability than the lipid ones and the compositions in amino acids did not show greater variations depending on the support. The results obtained by FTIR-ATR and CG-FID were analyzed applying multivariate statistical models, allowing a better interpretation of the data. In the second part of the work, we present the results obtained in the characterization of organic binders in wall paintings on plaster of two churches of 18th century of the Andean area, applying the analytical techniques and the optimized methodologies with the reference samples. The presence of vegetable oil and egg was identified as the binder of the pigments, which is characteristic of a tempera paint. We also detected the use of an animal glue as a primer for the gypsum preparation base. The identification of egg and collagen, a characteristic protein of animal glue, was confirmed by MALDI-TOF. These results acquired relevance when contrasted with information provided by historians and art restorers, constituting the first chemical study on pictorial technique in Andean mural painting.Fil: Castellanos Rodriguez, Diana Marcela.
