Effects of gamma-rays doses on shoot apex of banana (Musa sp.) plants developed in vitro aiming at mutation induction

Ápices caulinares de bananeira 'Maçã' foram tratados com raios gama (20, 40, 60, 80 e 100 Gy) e subcultivados in vitro por quatro ciclos vegetativos (geração M1V4), e concluiu-se, pelas variegações foliares observadas in vitro, que este é o número mínimo de gerações a ser utilizado antes da seleção de mutantes. Foram quantificadas, também, nesta geração, em casa de vegetação, as alterações fenotípicas (teor de antocianina, e alterações na forma, na variegação foliar e na arquitetura da planta), observando-se aumento das freqüências destes variantes com a elevação da dose. A dose de 40 Gy resultou em maior aumento da freqüência (13,49%) em comparação com o controle (0,85%), sendo a recomendada para trabalhos de indução de mutação com esta cultivar. Em casa de vegetação foram inoculadas, com suspensão de esporos de Fusarium oxysporum f. sp. cubense, agente causal do mal-do-panamá, 2.765 plantas jovens obtidas do controle e da geração M1V4. Não foram observadas plantas resistentes ao fungo, mas o sistema utilizado permitiu uma triagem rápida e eficiente das plantas suscetíveis.Shoot tips from banana cv. Based on leaf variegation observed in vitro it was concluded that MIV4 is the minimum number of generations to be used before the screening of mutants. At greenhouse, besides leaf variegation others phenotypic changes (anthocyanin, disturbed plant and leaf architecture) were also quantified. The frequency of this variants increased with the dose, and, the treatment with 40 Gy produced the highest frequency of 13,49% while in control plants it was only 0.85. So, this dose is recommended to be applied in mutation breeding with this cultivar. Inoculation of 2765 young plants were carried out in green-house with spores suspension of Fusarium oxysporum f. sp. cubense. No resistant plants were found, but this system permitted a quick and efficient screening of the susceptible ones and can be utilized for the selection of the mutants from big populations of banana cultivars
