Rheumatoid arthritis (RA) is a chronic inflammatory systemic disease that primarily affects the joints but may involve every organ of the human organism. RA has a prevalence of nearly 1% in the worldwide population. The disease is associated with significant disability and increased mortality if insufficiently treated. The pathogenesis of RA is not fully understood but is characterized by a strong genetic contribution and by abnormalities in the function of CD4 T cell subsets, which drive an adaptive autoimmune response. The cytokine interleukin 4 (IL-4), acting through the IL-4 receptor (IL-4R), crucially regulates CD4 T cell subset development and function, thereby contributing to RA development and/or progression.
