The significance of in vivo protein studies is widely appreciated. Whereas in vivo protein behaviours inside cells are influenced by many unique physical parameters, such as macromolecular crowding and intracellular viscosity, they are also affected by a plethora of biological activities that often vary greatly in different cell types. In-cell NMR spectroscopy is an invaluable biophysical tool for residue-specific, structural and functional investigations of isotope-labeled proteins inside native environments of cells and, as such, a preferred tool for studying intracellular proteins behaviours.
